摘 要:目的:优选大孔树脂分离纯化蜘蛛香总环烯醚萜类成分的工艺方法。方法:以Chlorovaltrate为对照品,采用紫外分光光度法测定蜘蛛香总环烯醚萜类成分的含量,动态吸附-解吸法筛选大孔树脂型号,并优选其纯化工艺条件。结果:D-101大孔树脂最适合蜘蛛香总环烯醚萜成分的分离纯化,最佳工艺为:上样液浓度为20g·L-1,用6 BV(Bed Volume)的水和4 BV的60%乙醇洗脱除杂,4 BV的95%乙醇洗脱,洗脱流速2 BV·h-1,收集95%乙醇洗脱液。洗脱液减压回收至干,在此工艺下所提取的蜘蛛香环烯醚萜类成分的纯度达66.3%。结论:D-101大孔树脂能有效提高蜘蛛香中总环烯醚萜类成分的纯度,工艺简单可行,较适宜应用于大生产。Objective : The aim of this study was to optimize the purification technology of total iridoid constituents from Valeriana jatamansi Jones. Method: The optimal macroporous resin was selected by dynamic adsorptiondesorption, and then the purification technology of total iridoid constituents from Valeriana jatamansi Jones was optimized with the iridoids content of Chlorovaltrate as index, which was determined by ultraviolet spectrophotometry. Result: The D -101 macroporous resin was the most suitable for purification of total iridoid constituents from Valeriana jatamansi Jones. The optimal purification conditions were:the mass concentration of sample liquid 20 g · L-1 ;the adsorption time 30 min, eluted by 6 BV water and 4 BV 60% ethanol to impurity;4 BV 95% ethanol, elution flow rate 2 BV · h-1 ;collected 95% ethanol eluent, and evaporated to dry. Under these technology conditions,the total iridoida constituent was up to 66.3 %. Conclusion:D - 101 type macroporous resin could effectively enhance purity of total iridoid constituents from Valeriana jatamansi Jones. This optimal technology was simple and feasible, which could meet the industrial requirements.
