摘 要:目的:探讨华蟾素注射液对H22肝癌小鼠肿瘤组织中Fas、FasL基因表达的影响。方法:40只ICR小鼠采用皮下接种H22肝癌细胞株的方法复制小鼠肝癌模型,造模成功后,将小鼠随机分为4组:模型组,顺铂组,华蟾素注射液低、高剂量组。采用荧光定量RT-PCR方法检测华蟾素注射液对H22肝癌小鼠肿瘤组织中Fas、FasL基因表达的影响。结果:华蟾素高剂量组、顺铂(DDP)组的抑瘤率分别为41.35%、53.85%,具有明显的抑制肿瘤生长作用。与模型组比较,华蟾素低剂量组小鼠肿瘤组织中的Fas、FasL mRNA表达无显著性差异(P>0.05)。而华蟾素高剂量组、DDP组能促进肝癌小鼠肿瘤组织中的Fas mRNA表达升高,与模型组比较有显著性差异(P<0.05),同时降低FasL mRNA表达,与模型组比较有显著性差异(P<0.05)。结论:华蟾素注射液抑制肝癌生长的分子机制可能是通过上调Fas基因表达并下调FasL基因表达,从而促进肿瘤细胞发生凋亡,达到抗肿瘤作用。Objective: To study the effect of Cinobufacini Injection on Fas and FasL gene expression in tumor tissues of mice bearing Hepatoma H22. Methods : Forty ICR mice were randomly divided into 4 groups : the model group, the Cisplatin group, the low and high dosage groups of Cinobufacini Injection after the mice model bearing Hepatoma H22 were established by subcutaneous inoculating tumor cells. The gene expressions of Fas and FasL in tumour tissues were detected by reverse -transcription polymerase chain reaction (RT -PCR). Results: The cancer inhibition ratio of the high dosage Cinobufacini Injection and Cisplatin group was 41.35% and 53.85% respectively and it showed obvious inhibitory effect on tumor growth. Compared with model group, the expression levels of Fas and FasL mRNA in tumor tissues from the mice bearing Hepatoma H22 treated with the low dosage Cinobufacini Injection showed no significantly difference in statistics( P 〉 0.05 ). Compared with the model group, the high dosage Cinobufacini Injection and Cisplatin group could promote the expression levels of Fas mRNA in mice tumor tissues of liver cancer and reduce the expression levels of FasL mRNA, and there was both significant difference in statistics( P 〈 0.05 ). Conclusions : Our results indicated that the molecular mechanism of inhibition of the liver cancer growth treated by Cinobufacini Injection may be through the up - regulating the expression levels of Fas gene and down - regulating the expression levels FasL gene so as to promote tumor cell apoptosis and achieve the anti -tumor effect.
