摘 要:目的:研究红景天苷(SDS)对百草枯(PQ)中毒大鼠肺组织Toll样受体4和核转录因子κB(TLR4-NF-κB)mRNA表达的影响。方法:将120只SD大鼠随机分为生理盐水(NS)对照组、PQ模型组和SDS干预组(低、中、高剂量),每组24只。采用一次性腹腔注射PQ溶液的方法,制备PQ中毒ALI模型,PQ溶液20 mg/kg。PQ模型组和SDS干预组注射PQ溶液,NS对照组腹腔注射等容积NS。染毒1 h后,SDS干预组腹腔注射SDS,低、中、高剂量分别为1、5和10 mg/kg,NS对照组、PQ模型组腹腔注射等容生理盐水,每24 h注射1次。分别在染毒后6 h、24 h、48 h和72 h麻醉处死大鼠,测定肺湿/干重比,测定肺组织中肿瘤坏死因子-α(TNF-α)、血清中白介素-6(IL-6)含量,检测TLR4和NF-κB的mRNA表达情况。结果:与NS组相比,各时间点PQ模型组肺湿/干重比明显增加(P<0.01),肺组织TLR4和NF-κB的mRNA表达水平明显提高(P<0.01),肺组织中TNF-α、血清中IL-6含量显著增加(P<0.01);而在SDS干预组中,与PQ模型组相比,中、高剂量组各时间点的肺湿/干重比明显减少(P<0.05),中、高剂量组在染毒后24、48、72 h肺组织TLR4和NF-κB的mRNA表达水平明显减少(P<0.05),各剂量组TNF-α的水平于染毒后各时间点均有显著降低(P<0.05)。高剂量组血清IL-6水平于染毒后6、24、48、72 h明显减低(P<0.05)。结论:TLR4-NF-κB通路参与了PQ中毒大鼠肺损伤的早期病理生理过程,SDS能抑制TLR4-NF-κB活化,减轻PQ中毒大鼠肺损伤。Objective: To observe the effects of salidroside (SDS) on (TLR4) mRNA expression and Nuclear Factor - KB( NF - KB) activity in the lung of paraquat poisoning rats. Methods: One hundred and twenty male SD rats were randomly divided into five groups ( each n = 24) : control group, model group, SDS intervention group ( including low, middie and high doses, 1,5,10 mg/kg, respectively). The ALI model induced by PQ poisoning was produced by intraperitoneal injection PQ solution (20 mg/kg), and the rats in control group were given normal saline solution with the same volume. One hour after the administration, rats in SDS intervention group were given SDS, and PQ group were given normal saline solution with the same volume, and injected once every 24 h. At 6,24,48 and 72 hours, the rats were sacrificed, and the lungs were taken away to determine the lung wet/dry weight ratios ( W/D ratios), the expressions of TLR4 mRNA, activities of NF - κB and levels of tumor necrosis factor - α (TNF - α) and Interleukin - 6 ( IL - 6) in the various groups. Results: The W/D ratios of lungs in rats of model group at 6,24,48,72 hours after poisoning were significantly higher than those in the normal control group, and compared with the model group, they were decreased significantly in the SDS intervention group ( middle and high doses). The expressions of TLR4 mRNA in the lung tissue of rats in model group at 6,24,48,72 hours after poisoning were markedly higher than those in the normal control group, while the TLR4 mRNA expression in SDS intervention group (middle and high doses) was markedly lower at 24,48,72 hours than that in the model group ( all P 〈 0.05 ) . The NF - κB activities in lung tissue of rats in model group at 6,24,48,72 hours after poisoning were obviously higher than those in normal control group, and compared with model group, the NF - κB activities were lower in SDS intervention group ( middle and high doses) at 24,48,72 hours ( all P 〈 0.05 ).
