益气活血方对球囊损伤大鼠颈总动脉后VEGF动态变化的影响及意义

摘　　要：目的:观察损伤血管内皮细胞生长因子(VEGF)的表达情况,探讨益气活血方对球囊损伤大鼠颈总动脉后再狭窄的干预机制。方法:随机将160只雄性SD大鼠分为假手术组,阴性对照组,益气活血方大、中、小剂量组。首先自左颈外动脉球囊损伤大鼠左颈总动脉,后给予高脂饲料喂养30 d,造成动脉硬化,然后经右股动脉穿刺球囊扩张损伤大鼠左颈总动脉,造成颈总动脉再狭窄。手术前1 d开始给药,每组动物于术后(3 d、7 d、14d、28 d)处死,取损伤颈总动脉,常规固定、包埋、切片,分别做HE染色和免疫组织化学SABC法检测VEGF的表达。结果:用药组VEGF表达明显增多,3 d、7 d和14 d益气活血方大剂量组VEGF表达量与阴性对照组、假手术组比较差异有显著性(P<0.05)。结论:益气活血方对球囊损伤后大鼠颈动脉再狭窄有明显的防治作用,其机制可能与上调VEGF的表达,促进血管内皮细胞修复与新生有关。Objective： To investigate the significance of Yiqi Huoxue prescriptions on abnormal expression of VEGF protein after carotid artery vascular injury in rats. Methods： Rats （n = 160）were randomly divided into 5 groups：the sham - operated group ; the model group ; the low - dose Yiqi Huoxue group, the mid - dose Yiqi Huoxue group and the high - dose Yiqi Huoxue group. In the latter 4 groups, the rats＇ left common carotid artery was injured by balloon through the left external carotid artery. And then all of the rats were fed up with hypercholesterolemic diet for 30 days. After that, the 2. OF balloon catheter injury was performed through the right femoral artery into the left common carotid artery of rats and the balloon was inflated and deflated for 3 times. The rats were given drugs into stomach from 1 day before injury and to 3 days, 7 day% 14 days ,28 days after injury. Around 8 rats from each group were euthanized by exsanguination at appropriate time point, the injured left common carotid arteries was carefully removed and harvested for HE staining and SABC staining. The expression of VEGF protein was observed by immunohistochemical technique. Results：The expression of VEGF proteinat the 3rd day,the 7th day and the 14th day after injury was increased obviously when given Huoxue Yiqi prescriptions compared with those of the model group and the sham - operated group （ P 〈 0. 05 ） . Conclusion ： Huoxue Yiqi prescriptions in the prevention and treatment of RS after balloon injury might be related to promoting the secretion of VEGF protein, and thus accelerating reendothelialization.
