摘 要:目的:观察软肾散对单侧输尿管梗阻(UUO)大鼠模型肾间质纤维化(RIF)的影响及可能的作用机制。方法:将48只SD大鼠随机分为假手术组、模型组、依那普利治疗组和软肾散治疗组,后3组建立UUO大鼠模型。治疗结束后测定大鼠肾功能、行HE染色观察肾组织病理变化,计算肾小管间质病理积分,用免疫组化方法测定肾TGF-β1、α-SMA的表达情况。结果:两治疗组大鼠BUN、SCr低于模型组(P<0.01),两治疗组组间差异无统计学意义(P>0.05);肾间质病理积分显示两治疗组显著低于模型组(P<0.01),两治疗组组间差异无统计学意义(P>0.05);免疫组化结果表明,两治疗组TGF-β1和α-SMA的表达均低于模型组(P<0.01),两治疗组组间差异无统计学意义(P>0.05)。结论:软肾散能一定程度上减轻UUO大鼠RIF程度,其作用机制可能与下调TGF-β1和α-SMA表达有关。Objective : To observe the therapeutic effect of Ruamshen Powder on the renal interstitial fibrosis of unilateral ureteral obstruction (UUO) rat model and then to study its possible mechanism. Methods : Forty - eight Sprague - Dawley male rats were randomly divided into four groups : sham - operation group, model group, Emalapril group and Ruamshen Powder treatment group. After operation,we tested the serum creatinine (SCr), blood urea nitrogen (BUN). At the same time, the pathological examine were performed, and the renal interstitial pathology points were evaluated. The expression levels of TGF - β1 (transforming growth factor - β1 ) , α - SMA (α - smooth muscle actin) were detected by immunohistochemistry. Results :The level of renal function in UUO group was obviously higher than that in Ruamshen Powder treatment group and Emalapril group (P 〈 0.01 ). And there were no significant differences between two treatment groups (P 〉 0.05 ). Immunohistochemistry showed that the expression level of TGF - β1 and α - SMA in Ruamshen Powder treatment group and Emalapril group were lower than that in UUO group after operation ( P 〈 0.01 ) and there were no significant differences between two treatment groups ( P 〉 0.05 ). Conclusion : Ruamshen Powder can partly delay the progress of renal interstitial fibrosis in UUO rat model. Its mechanism may be related to suppressing the expression of TGF - β1 and α - SMA.
