摘 要:〔目的与方法〕采用 5 1 Cr释放法对 MTT比色法及 NAG释放法测定恶性肿瘤患者 NK细胞介导的细胞毒活性的价值进行初步探讨。结果 :(1)两种非同位素方法所测外周血 NK细胞活性与5 1 Cr释放法之间均无相关性 (P均 >0 .0 5 ) ;(2 )正常对照组效应细胞 5 1 Cr释放水平低于靶细胞 (P<0 .0 1) ,肿瘤化疗组效应细胞5 1 Cr释放水平则高于靶细胞(P<0 .0 1) ,且靶细胞与效应细胞的结果无相关性 (P>0 .0 5 )。〔结论〕我们认为酶反应比色法不宜用于 NK细胞介导的细胞毒活性测定。睴bjective〕〔Methods〕: 51 Cr release,MTT and NAG assays were performed in cancer patients to evaluate the colorimetric assay basing on enzymatic reaction in NK cell-mediated cytotoxicity. 〔Results〕:(1)There was no correlation of NK cytotoxicity values between 51 Cr release and the colorimetric assays in cancer patients(P>0 05);(2)In normal control group the levels of 51 Cr release from effector cells were lower than those from target cells(P<0 01),but in chemotherapy group the former higher than the latter(P<0 01)and no linear relationship of the levels of 51 Cr release was found between effector and target cells(P>0 05).〔Conclusions〕:We believed that the colorimetric assay basing on enzyme reaction is not suitable to NK cell-mediated cytotoxicity.
