摘 要:目的探讨CRIF1基因对L615白血病细胞小鼠体内增殖的影响。方法实验分为3组,CRIF1-RNAi L615实验组(n=3):慢病毒干扰载体抑制L615细胞CRIF1-GFP基因表达;空载体对照组(n=3):空载体转染L615细胞;空白对照组(L615细胞空白对照)(n=3);建立白血病小鼠模型。通过肝脾印片、骨髓涂片及外周血流式细胞仪检测观察各组小鼠体内L615细胞增殖情况,比较各组小鼠的外周血白细胞计数(WBC)、生活质量和生存时间、肝脾指数。结果流式细胞仪检测:CRIF1-RNAi L615实验组、空载体对照组的小鼠外周血均见大量GFP阳性L615细胞;肝、脾印片及骨髓涂片结果显示肝、脾、骨髓均被白血病细胞浸润,3组小鼠均成瘤。CRIF1-RNAi L615实验组、空载体对照组及L615对照组小鼠在白血病细胞植入d5后外周血WBC(×109/L)分别为:42.23±15.08 vs 17.10±4.10 vs 17.30±5.05(P<0.01);生存时间(d)分别为:6.33±1.15 vs 10.00 vs 8.33±2.89(P<0.01);CRIF1-RNAi L615实验组小鼠脱毛、弓背等并发症出现提前;肝、脾指数分别为:122.31±2.83、35.21±0.45 vs 110.89±13.30、21.54±3.40 vs 89.76±5.56、24.71±2.48(P<0.05)。结论抑制小鼠L615白血病细胞CRIF1基因表达后,明显著促进L615白血病细胞在小鼠体内增殖、浸润并加快移植小鼠发病死亡。Objective To study the impact of CRIF1 gene on the proliferation of L615 leukemia cells in mice . Methods Three groups of L615 cells ( L615 cells with lentiviral vectors interference suppressing CRIF1-GFP gene;L615 cells with empty vector;And control L615 ceils)were used to establish the mouse model of leukemia, and the proliferation of L615 cells in each group were observed through imprints of liver and spleen, bone marrow aspirate or flow cytometry of peripheral blood. Then we compared the WBC counts of peripheral blood, quality of life, survival time, index of liver and .spleen in each groups mice. Results A large number of GFP-positive L615 cells were observed in peripheral blood both groups of Crifl-RNAi L615 and empty vector mice by flow eytometry. Leukemia cells in liver, spleen, bone marrow were identified, and tumorigenesis were observed in all groups. And the peripheral WBC counts( x 109/L) in Crifl-RNAi L6i5 group ,empty vector mice and L615 mice since the fifth day of injection of leukemia cells were 42. 23 ± 15.08 vs 17. 10 ± 4. 10 vs 17. 30 ±5.05 ( P 〈 0.01 ), with significantly shorter survival time ( d ) were 6. 33 ±1.15 vs 10. 00 vs 8.33± 2. 89 ( P 〈 0. 01 ), advance in hair removal, arched shoulders . The liver and spleen index were 122. 31± 2, 83,35. 21 ± 0.45 vs 110. 89 ± 13.30,21.54 ±3.40 vs 89. 76± 5.56,24. 71± 2.48 ( P 〈0. 05 ). Conclusion The inhibition of Crifl gene expression significantly promotes the proliferation of murine L615 leukemia cells in vivo and accelerates the pathological process of leukemia.
