摘  要:目的比较5种标本浓缩方法处理标本前后ELISA检测HBsAg的灵敏度。确定经济、适用的标本浓缩方法。方法采用聚丙烯酰胺凝胶吸附法、聚乙二醇透析法、高速离心沉淀法、磁珠浓缩法、冷冻干燥法分别浓缩HBsAg阳性血清标本,比较浓缩前后的S/CO值。结果浓缩后的S/CO值均高于浓缩前(P<0.01),磁珠浓缩法浓缩能力高于聚丙烯酰胺凝胶吸附法、聚乙二醇透析法和高速离心法(P<0.05)。结论标本浓缩法可以提高ELISA检测HBsAg的灵敏度,磁珠浓缩法具有明显的优势,很适合在临床上推广使用。Objective To compare the sensitivity of ELISA for HBsAg detection which dealt with five enrichment methods for serum samples before and afterwards ; and to determine the most economic and effect enrichment method. Methods Five enrichment methods including polyethylene glycol (PEG) dialysis, polyacrylamide gel particle adsorption, high-speed centrifugal, magnetic beads enrichment method, and vacuum dry deal were performed on the serum samples, the S/CO values before and after concentration were compared. Results The S/CO values of the all concentrated samples were higher than that of before ( P 〈 0. 01 ). The concentration ability of magnetic beads enrichment method was higher than polyacrylamide gel particle adsorption,polyethylene glycol (PEG) dialysis and high-speed centrifugal (P 〈 0. 05 ). Conclusion Enrichment methods can obviously improve the sensitivity of ELISA for the detection of HBsAg, Megnetic beads enrichment method has the obvious advantage comparing to the 'other concentration methods, and it is suggested to be used widely in clinic.
