摘 要:目的比较大车前草及车前草中大车前苷、总黄酮的含量差异。方法采用HPLC法测定大车前苷,色谱柱为Dikma Diamonsil C18(250mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸(18:82),流速1mL·min^-1,检测波长330nm,柱温30℃。以70%乙醇为提取溶剂,以木犀草素为对照品;采用紫外-可见分光光度法测定总黄酮。结果大车前苷0.09~11.22μg与峰面积呈线性(r=0.999),加样回收率为100.64%(RSD=0.95%),测得4批大车前草中大车前苷的含量为0.09%~1.36%,6批车前草中大车前苷的含量为0.14%~2.40%。总黄酮4.248—50.976μg·mL^-1与吸光度呈线性(r=0.999),加样回收率为100.13%(RSD=1.55%),测得4批大车前草中总黄酮含量为1.03%~4.86%,6批车前草中总黄酮的含量为1.31%-4.98%。结论两种车前草中大车前苷的含量有一定差异,总黄酮含量较高且差异不大,这可能是导致其药效差异的物质基础。OBJECTIVE To compare the content of plantamajoside and total flavanone in Plantago major L. and Plantago asiatica L. METHODS An HPLC method was established to determine the content of plantamajoside. Plantamajoside was separated at 30℃ on a Dikma Diamonsil C18(250 mmx 4.6 mm, 5μm) column with acetonitrile - 0. 1% phosphoric acid ( 18 : 82) as the mobile phase. The detection wavelength was set at 330 nm and the flow rate was 1 mL·min^-1. With 70% ethanol as the extraction solvent and luteolin as the reference substance,the total flavanone was determined by the ultraviolet spectrophotometry. RESULTS In the HPLC determination, good linearity of the calibration curve between the peak area and the concentration of plantamajoside was obtained in the range of 0.09 - 11.22μg( r = 0. 999), and the average recovery of plantamajoside was 100.64% with RSD of 0.95%. The contents of plantamajoside were in the range of 0.09% - 1.36% in 4 batches of Plantago major L. while 0.14% - 2.40% in 6 batches of Plantago asiatica L. In the ultraviolet spectrophotometry determination, good linearity of the calibration curve between the absorbency and the concentration of total flavanone was obtained in the range of 4. 248 -50. 976μg·mL^-1 (r = 0. 999 ), and the average recovery of total flavanone was 100.13% with a RSD of 1.55%. The contents of total flavanone were in the range of 1.03% -4.86% in the Plantago major L. while 1.31% -4.98% in Plantago asiatica L. CONCLUSION The content of plantamajoside in the two samples are different while the content of total flavanone are similar, it may be the reason of the different pharmacodynamics activities and can be further developed and used,
