摘  要:目的探讨高翅果菊提取物莴苣苷B对缺血性脑卒中大鼠海马中VEGF mRNA及其蛋白表达的影响。方法将SD大鼠随机分为6组,即假手术组、模型组、阳性对照组、莴苣苷B低、中、高(12.5、25、50 mg·kg-1)剂量组,大鼠均按5 mL·kg-1ip给药,bid。各组于给药24 h后处死7只小鼠,其中4只用于免疫组化研究,3只用于RT-PCR研究,剩余小鼠于给药72 h后处死,每组的4只用于免疫组化研究,其他用于RT-PCR研究。结果再灌注24 h时模型组海马中VEGF的基因和蛋白表达增多,与假手术组比较有显著性差异(P<0.01),但再灌注72 h时VEGF基因表达有所下降。用药后,莴苣苷B各剂量组小鼠的VEGF mRNA和蛋白表达均增多,与模型组比较有显著性差异(P<0.05,P<0.01),其中,以给药72 h的中、高剂量组的作用更明显,与模型组和阳性对照组比较均有显著性差异(P<0.05,P<0.01)。结论莴苣苷B可能通过上调缺血性脑卒中大鼠海马中VEGF基因和蛋白的表达,促进缺血半暗区或海马敏感区早期的神经发生和后期的微循环形成而改善缺血性脑卒中。OBJECTIVE To explore the effect of extractive lactuside B ( LacB ) from Pterocypsela elata on the expression of hippocampus VEGF mRNA and protein in the ischemic stroke rats. METHODS SD male rats were randomly divided into six groups:Sham operation group, model group, positive group, LaeB low dose ( 12.5 mg·kg - 1 ) group, LacB middle dose ( 25 mg·kg - 1 ) group and LacB high dose( 50 mg·kg-1 ) group. All the rats received intra -peritoneal injections twice a day with a dosage of 5 mL·kg-1. Seven animals from each group were sacrificed at 24 h, four rats were used for detection of VEGF protein expressions in hippocampus by immunohistochemistry method, and three rats were used for detection of VEGF mRNA expressions by RT - PCR technique. The rest animals were sacrificed at 72 h after giving medications, four rats were used for immunohistoehemistry technology, and others were used for RT - PCR. RESULTS After repeffusion for 24 h, model group VEGF gene and protein expression increased significantly compared with the sham group in the hippoeampus ( P 〈 0.01 ) , but after reperfusion for 72 h VEGF mRNA and protein expression decreased. VEGF mRNA and protein expression of all the LacB groups increased after giving drugs, which was significantly different from that of model group( P 〈 0. 05 ,P 〈 0.01 ). 72 h after drug administration,VEGF expression of medium and high dose group had more VEGF expression, while those of the model group and positive drug group were significantly different ( P 〈 0.05,P 〈 0.01 ). CONCLUSION LacB could ameliorate ischemic stroke through promoting early neurogenesis and formation of the late microcireulation in ischemic penumbra and hippocampal sensitive area, which may be attributed to its VEGF expression induction ability in the hippocampus.
