摘  要:目的采用反相HPLC法同时测定中亚沙棘中的原儿茶酸、芦丁、异鼠李素。方法采用Gemini C18色谱柱(150mm×4.6mm,5μm),柱温25℃,流动相为甲醇-0.05%磷酸梯度洗脱(5:95→40:60→60:40),流速1.0mL·min^-1,检测波长256nm。结果中亚沙棘中原儿茶酸0.019~0,808μg(r=0.9998)、芦丁0.022~0.823μg(r=0.9998)、异鼠李素0.014—0.443μg(r=0.9999)与峰面积的线性关系良好;三者的平均加样回收率(n=9)分别为98.57%(RSD=1.57%)、99.13%(RSD=1.17%)、96.22%(RSD=1.81%)。结论所用方法简单、快速、高效,可为中亚沙棘的质量控制提供依据。OBJECTIVE To establish an RP - HPLC method for the simultaneous determination of protocatechuic acid, rutin and isorhamnetin in Hippophae rhamnoides L. subsp, turkestanica Rousi in. Ann.. METHODS The determination was carried out on a Gemini C18 column( 150 mm ×4.6 mm,5μm) with the column temperature at 25℃. The mobile phase was a mixture of methanol and 0.05% phosphoric acid water solution with the gradient elution(5 : 95→40 : 60→60 : 40). The flow rate was 1.0 mL·min^-1 and the detection wavelength was 256 nm. RESULTS The method showed a good linear relationship within the range of 0. 019 -0. 808μg for protocateehuic acid ( r = 0.9998 ) ,0. 022 - 0. 823 μg for rutin ( r = 0. 9998 ) and 0.014 - 0. 443μg for isorhamnetin ( r = 0. 9999 ). The average recoveries(n =9) were 98.57% with RSD of 1.57% ,99.13% with a RSD of 1.17% ,and 96.22 % with a RSD of 1.81% , respectively. CONCLUSION The method is simple, rapid and efficient, which provides the basis for quality control of H. rhamnoides.
