摘  要:目的:建立以HPLC多波长梯度洗脱法同时测定达瓦依金丸中的龙胆苦苷、胡椒碱、木香烃内酯、去氢木香烃内酯、芦荟大黄素、大黄素、大黄酚、大黄素甲醚8个活性成分。方法:采用Waters C18(250mm×4.6mm,5μm)色谱柱;乙腈-0.1%磷酸溶液为流动相梯度洗脱,运行时间45min,平衡时间为15min;流速1.0mL·min^-1;柱温为25℃;在270nm处检测龙胆苦苷、在225nm处检测木香烃内酯和去氢木香烃内酯、在254nm处检测芦荟大黄素、大黄素、大黄酚、大黄素甲醚和343nm处检测胡椒碱。结果:龙胆苦苷、胡椒碱、木香烃内酯、去氢木香烃内酯、芦荟大黄素、大黄素、大黄酚、大黄素甲醚分别在35.8~357.9μg·mL^-1(r:0.9993),49.9~498.8μg·mL^-1(r=0.9999),12.0~119.7μg·mL^-1(r=0.9993),16.8~168.3μg·mL^-1(r=0.9995),5.6~56.2μg·mL^-1(r=0.9996),7.4~74.0μg·mL^-1(r=0.9998),15.7~156.8μg·mL^-1(r=0.9991),5.7~57.4μg·mL^-1(r=0.9999)范围内与峰面积呈良好的线性关系;平均加样回收率(n=6)分别为:98.93%,97.96%,99.38%,99.22%,99.12%,97.37%,98.94%和99.03%,RSD分别为1.65%,1.98%,1.99%,1.53%,1.23%,1.67%,2.10%和0.44%(n=6)。结论:该方法简便、准确、重复性好,可为达瓦依金丸的质量控制提供实验依据。Objective :To establish a multi-wavelength HPLC method for simultaneous determination of gentiopicroside,piperine, costunolide, dehydrocostus-lactone, alon-emodin, emotion, chrysophanol and physcion in Uighur medicine Dawayijin pills. Methods: Chromatographic separation was performed on a Waters Cls column (250 mm×4.6mm,5μm)maintained at 25 ℃. The mobile phase was acetonitrile-0.1% phosphoric acid eluted gradiently at a rate of 1.0 mL·min^- 1 The detection wavelengths were 270 nm(gentiopicroside) ,225 nm(costunol- ide and dehydrocostuslactone) ,254 nm ( alon-emodin, chrysophanol, emodin and physcion) and 343 nm (piperine). The running time was 45 minutes,and balance time was 15 minutes. Results:The linear ranges were 35.8 -357.9μg·mL^-1 ( r = 0. 999 3 ) for gentiopicroside, 49.9 - 498.8μg·mL^-1( r = 0. 999 9 ) for piperine, 12.0 - 119.7μg·mL^-1( r = 0. 999 3 ) for eostunolide, 16.8 - 168.3μg·mL^-1 ( r = 0. 999 5 ) for dehydrocostus-lactone, 5.6 -56.2μg·mL^-1 ( r = 0. 999 6) for alon-emodin,7.4 - 74.0 μg·mL^-1( r = 0. 999 8 ) for emotion, 15.7 - 156.8μg·mL^-1( r = 0. 999 1 ) for ehrysophanol, and 5.7 - 57.4μg·mL^-1(n=0. 999 9 ) for physcion, respectively. The average recoveries( n = 6) of the eight components were 98.93% ,97.96% ,99.38% ,99.22% ,99.12% ,97.37% , 98.94% and 99.03% ,with RSDs of1. 65% ,1. 98% ,1. 99% ,1. 53% ,1. 23% ,1. 67% ,2.10% and 0.44%(n= 6) ,respectively. Conclusion:The developed method is convenient, rapid and accurate with high repeatability and stability,which is suitable for the content determination of Dawayijin pills.
