摘 要:目的:研究内质网应激蛋白在肾纤维化大鼠肾组织的表达及水苏碱的干预作用。方法:采用单侧输尿管梗阻诱导大鼠肾间质纤维化的动物模型,将大鼠分为假手术组、模型组、依那普利组、水苏碱93,46,23 mg·kg-1·d-1剂量组;采用HE,Masson染色检测肾小管损伤指数及肾间质纤维化程度;TUNEL法检验肾小管上皮细胞凋亡情况;免疫组织化学法检测大鼠肾组织GRP78,GRP94的蛋白表达。结果:水苏碱各治疗组与模型组比较上述指标均有统计学上的显著性差异(P<0.05或0.01),水苏碱93 mg·kg-1·d-1与依那普利组比较有差异(P<0.05)。结论:水苏碱可能通过上调ERS蛋白GRP78,GRP94的表达,阻碍肾小管上皮细胞凋亡,从而减轻肾间质纤维化的进展。Objective: To observe the expression of endoplasmic reticulum stress protein in renal tissue of rats with renal interstitial fibrosis and the intervention effect of stachydrine. Methods: Unilateral ureter obstruction (UUO) was used to induce rat renal interstitial fibrosis (RIF) model. Rats were divided into 6 groups, including sham operation group, model group, enalaprilat group, and stachydrine 93,46, 23 mg·kg^-1·d^-1 groups. HE and Masson methods were used to detect tubular injury index and degree of RIF. TUNEL was used to detect tubular epithelial cell apoptosis. The expressions of GRP78 and GRP94 in renal tissue were detected with immunohistochemical method. Results: The above indicators had statistically significant difference between the model group and the stachydrine treatment groups (P 〈 0.05 or 0.01 ). Compared with the enalapril group, the above indicators were different in stachydrine 93 mg·kg^-1·d^-1 dosage group ( P 〈 0.05 ). Conclusion : Stachydrine might increase the expressions of ERS protein GRP78 and GRP94. Tubular epithelial apoptosis was obstructed to reduce the progress of RIF.
