摘  要:目的 探讨IL 12反义寡核苷酸对狼疮肾炎 (LN)患者单个核细胞 (PBMC)分泌免疫球蛋白的影响。方法 采用反义寡核苷酸技术 ,应用ELISA法观察培养的PBMC上清液中IL 12及IgG的水平变化。结果 IL 12p40反义寡核苷酸 (ASODN)可明显抑制活动期LN患者PBMC自发分泌IL 12 ,且在一定的范围内呈剂量 (0~ 8μmol/L)与时间 (0~ 5h)依赖性 ;IL 12 p40ASODN抑制活动期LN患者PBMC自发分泌IgG ,在一定浓度范围内 (0~ 8μmol/L)呈剂量依赖性 ,而正义寡聚核苷酸 (SODN)与随意寡聚核苷酸 (RODN)则无这种抑制作用。结论 IL 12p40ASODN能有效抑制活动期LN患者PBMC体外分泌IL 12 p70、IgG ,有潜在的应用前景。Objective To investigate the effect of IL 12 antisense oligonucleotides on immunoglobulin secretion by PBMC from patients with active lupus nephritis (LN).Methods Levels of IL 12 p70 and IgG in the supernantants of PBMC treated with IL 12 p40 ASODN were measured by ELISA technique.Results IL 12 p40 ASODN inhibited evidently spontaneous IL 12 production by PBMC from active LN patients in a time dependent (0~5 hours) and a dose dependent (0~8 μmol/L) manner.IL 12 ASODN inhibited markedly spontaneous IgG production by PBMC from active LN patients in a dose dependent (0~8 μmol/L) manner.Whereas IL 12 p40 SODN and RODN had no effect.Conclusion DOATP Lipo IL 12 p40 ASODN complex made by the authors of this article definitely inhibits spontaneously IL 12 p70 and IgG antibody production by PBMC from active LN patients in vitro with good stability and permeability,and it is potentially used as a medicine for active lupus nephritis.
