摘  要:目的:探讨不同血样处理方法对人类免疫缺陷病毒1型(HIV-1)病毒载量检测结果的影响,以提高检测准确率。方法采用H IV-1核酸序列扩增技术对不同方法处理后的样品进行检测,主要观察条件为离心、血浆样品保存温度、保存时间以及冻融次数等。结果不同血样处理方法基线样品的检测值误差为0.00114/lg ,在可控范围内(病毒载量检测值变化<0.2/l g );未经分离血浆的检测结果为0.219~0.444/lg与基线值比较差异有统计学意义(病毒载量检测值变化>0.2/lg );对抗凝后的血液样品进行适当的分离,将有助于更加准确地检测病毒载量,对于离心后的血浆样品,其最佳保存条件为室温下6 h、-4℃条件下可保存至24 h ,冻融次数以-20℃冻融1次为佳、-70℃可冻融2次后即检测。结论离心、低温和适当冻融将有助于提高 HIV-1病毒载量检测的准确度。OBJECTIVE To explore the effects of different blood processing methods on human immunodeficiency virus 1(HIV-1) viral load test results ,so as to improve the detection accuracy .METHODS The HIV-1nucleic acid sequence-based amplification technology was adopted to test the samples processed with different methods ;the major observation conditions were composed of storage temperature ,storage time and freezing and thawing times for centrifuged plasma samples .RESULTS The deviation from detection value of the baseline samples treated with different methods was 0 .00114/lg ,within the scope of control(the change of viral load detection value was less than 0 .2/lg);the detection result for the plasma without separation varied from 0 .219 to 0 .444/lg ,comparing with the baseline value ,there was significant difference (the change of viral load detection value was larger than 0 .2/lg);the appropriate separation of blood sample after anticoagulation could contribute to more accurate detection of viral load .For plasma samples after centrifugation ,the optimum storage conditions were :6 hours under room temperature ,and 24 hours under -4 ℃ temperature;once freezing and thawing under -20 ℃ was the best condition and the blood sample could be detected after twice freezing and thawing under -70 ℃ . CONCLUSIONS Centrifuging ,low temperatures and appropriate freeze -thaw would help to improve the detection accuracy of HIV-1 viral load .
