LY294002对糖尿病肾病大鼠肾组织IL-1β与IL-6表达的影响

摘　　要：目的研究经静脉注射PI3K抑制剂LY294002对糖尿病肾病(DN)大鼠模型肾组织白介素-1β(IL-1β)与白介素-6(IL-6)表达的影响。方法成年健康清洁级SD雄性大鼠30只,随机分为假手术组(10)、模型组组(10)和PI3K抑制剂(LY294002)处理组。参照Anderson's方法制作糖尿病肾病大鼠模型。采用Western blot方法检测三组大鼠肾组织IL-1β与IL-6蛋白的表达;RT-PCR方法检测三组大鼠肾组织IL-1βmRNA与IL-6mRNA的表达。结果 Western blot结果显示,模型组与LY294002处理组肾组织IL-1β与IL-6蛋白的表达量均显著高于假手术组(<0.05),而LY294002处理组比模型组肾组织IL-1β与IL-6蛋白的表达量明显降低(<0.05)。RT-PCR结果显示,与假手术组比较,模型组与LY294002处理组肾组织IL-1βmRNA与IL-6mRNA的表达量均显著升高(<0.05),而LY294002处理组肾组织IL-1βmRNA与IL-6 mRNA的表达则明显低于模型组(<0.05)。结论 PI3K抑制剂LY294002能够下调DN大鼠肾组织IL-1β与IL-6的表达。Objective To study the effect of LY294002 on the expressions of intedeukin -1β （IL-1β ） and interleukin -6（IL-6） in kidney of the diabetic nephopathy rat. Methods 30 healthy male rats were selected and randomly divided into the sham group（10）,the model group（10） and LY294002 treatment group. The diabetic nephropathy model was induced with Anderson＇s method. The expressions of IL-1β and IL-6 were observed by Western blot method, the expressions of IL-1β mRNA and IL-6 mRNA were observed by RT-PCR method. Resdts The expression levels of IL-1β and IL-6 proteins and mRNAs were increased significantly in model group and LY294002 treatment group than in sham group（P〈0.05）, but decreased in LY294002 treatment group than in model group （P〈0.05）. Conclusion LY294002 could down-regulate the expressions of IL-1β and IL-6 in kidney of the diabetic nephopathy rat.
