摘  要:目的研究紫草素对脂多糖诱导的新生大鼠急性肺损伤(ALI)的保护作用。方法 7d龄SD大鼠40只随机均分为四组,采用脂多糖5mg/kg腹腔注射建立ALI大鼠模型。A、B和C三组分别于建模前1h用紫草素50、25和12.5mg/kg腹腔注射,D组为模型对照。另取10只7d龄SD大鼠作为空白对照(E)组。6h后,用ELISA测定各组大鼠血清TNF-α和IL-10的表达,并获取肺组织,测定肺组织湿干重(W/D)比率,观察病理变化。结果与D组比较,紫草素预处理组大鼠ALI呈剂量依赖性减轻,肺组织W/D比率改善(P<0.05),抑制TNF-α表达,上调IL-10表达(P<0.05或P<0.01)。结论紫草素预处理对模型大鼠ALI具有保护作用,可能与抑制TNF-α表达和增加IL-10表达有关。Objective To study the protective effects of shikonin in lipopolysaccharide(LPS)induced acute lung injury(ALI) of neonatal rats. Methods Forty 7-day neonatal SD rats models were established by intraperitoneal injection of LPS 5 mg/kg,which were equally randomized into 4 groups and treated with different dosage of shikonin 50 mg/kg (group A), 25 mg/kg (group B) and 12. 5 mg/kg(group C) at one hour before LPS injection. Normal saline was used in group D as model control. Another 10 7-day neonatal SD rats were taken as blank control(group E). At 6 hours after LPS injection, serum expressions of TNF-α and IL-10 were detected by ELISA and lung tissues were taken for pathological observation and calculation of the ratio of lung wet to dry weight(W/D). Results Compared to group D, ALI in groups of A, B and C was significantly improved dose-dependently with reduced W/D ratio(P〈0. 05),suppressed TNF-α expression and elevated IL-10 expression(P〈0. 05 or P〈0. 01). Conclusion Shikonin pretreatment has a protective effect against LPS-induced ALI of neonatal rats,which may be related with a decrease of TNF-α expression and an increase of IL-10 expression.
