七氟醚对福尔马林致炎性痛大鼠脊髓的p-CREB表达的作用

摘　　要：目的:初步探讨七氟醚对福尔马林诱发的急性炎性痛大鼠脊髓内p-CREB表达的作用。方法:通过足底皮下注射福尔马林建立大鼠急性炎性痛模型,采用免疫组化的方法,观察七氟醚吸入对福尔马林炎性痛引起的大鼠脊髓内环腺苷酸反应元件结合蛋白(cAMP response element binding protein,CREB)磷酸化的影响。结果:大鼠足底皮下注射福尔马林后,10 min内脊髓内p-CREB(磷酸化CREB)表达相比对照组明显增加并达到峰值(P<0.001),1 h时下降,但仍高于正常水平(P<0.001);短时间吸入七氟醚不能抑制注射福尔马林后10 min的脊髓p-CREB表达(P>0.05),但长时间吸入能完全抑制注射后1 h的脊髓内p-CREB表达(P<0.001)。结论:长时间吸入七氟醚对福尔马林炎性痛引起的脊髓p-CREB的表达有抑制作用,提示长时间吸入七氟醚对福尔马林炎性痛可能具有镇痛作用,且可能机制是通过调节脊髓内p-CREB的表达。Objective To explore the effect of sevoflurane on expression of p-CREB in spinal cord of rats with acute in-flammatory pain induced by formalin. Methods： Immunohistoehemistry was used to observe the effect of sevoflurane onphosphorylation of CREB （cAMP response element binding protein） in spinal cord induced by acute inflammatory pain inrat model which was produced by intra-plantar subcutaneous injection of formalin. Results ： After intraplantar injection offormalin, the expression of p-CREB in spinal cord was increased dramatically and reached to the peak within 10 min （P〈0. 001 ）, and then decreased but still higher than the control level at 1 h post-injection （P 〈0. 001）; long-termsevoflurane anesthesia inhibited the expression of p-CREB in spinal cord at 1 h post-injection of formalin （ P 〈 0. 001 ）,but transitory sevoflurane anesthesia did not inhibit at 10 min post-injection of formalin （P 〉 0.05 ）. Conclusion： Long-term sevoflurane anesthesia has suppressive effect on expression of p-CREB in spinal cord of rats activated by formalin-in-duced inflammatory pain, which suggests that long-term sevoflurane anesthesia may play analgesia effect on inflammatorypain induced by formalin with regulating the expression of p-CREB in spinal cord.
