摘 要:目的研究半乳糖凝集素-3(galectin-3,Gal-3)在上皮性卵巢癌中的表达及对人上皮性卵巢癌细胞株增殖、迁移和侵袭能力的影响。方法分别采用real-lime PCR和Western blot法检测上皮性卵巢癌中Gal-3的mRNA及蛋白表达;在上皮性卵巢癌细胞中运用小干扰RNA(small interfering RNA,siRNA)和慢病毒感染方法,分别下调和过表达Gal-3后,采用CCK一8试剂盒检测细胞的增殖,Matrigei和Transwell小室检测细胞的迁移及侵袭能力,蛋白质印迹法检测细胞周期蛋白D1(Cyclin D1)、基质金属蛋白酶-9(MMP-9)和E-钙黏蛋白(E-cadherin)的蛋白表达水平。结果与正常卵巢组织相比,Gal-3在上皮性卵巢癌组织中高表达,并与临床分期有关(P<0.05)。在上皮性卵巢癌细胞中分别过表达和下调Gal-3后,癌细胞的增殖、迁移和侵袭能力均相应地增强和下降,与对照组比较均有明显统计学意义(P<0.001),Cyclin D1和MMP-9蛋白表达分别上调和下调,而E-钙黏蛋白表达分别出现下调和上调。结论 Gal-3在上皮性卵巢癌中高表达,并促进上皮性卵巢癌细胞的增殖、迁移及侵袭。Objective To investigate the expression of Gal-3 in epithelial ovarian cancer(EOC) and its effects on proliferation,migration and invasion in EOC cells.Methods We detected the mRNA and protein expression of Gal-3 in the in EOC tissue specimens by real-time polymerase chain reaction (PCR) and Western blot respectively.After siRNA-mediated knockdown of Gal-3 or lentivirus-mediated overexpression of Gal-3 in EOC cell lines,CCK-8 kits were used to detect the proliferation in EOC cells.Matrigel and transwell were used to observe cellular migration and invasion ability.Western blot were used to detect the protein expression levels of Cyclin D1,matrix metalloproteinase-9 (MMP-9) and E-cadherin.Results Compared with normal ovarian tissues,Gal-3 in EOC was highly expressed and correlated with clinical stage (P<0.05).The capacity of proliferation,migration,invasion of EOC cell with down-regulation Gal-3 were decreased,the difference among them and control group was statistically significant (P<0.001),the protein expression levels of Cyclin D1 and MMP-9 were decreased,the protein expression level of E-cadherin was increased;overexpression of Gal-3 increased cell proliferation,migration and invasion,the difference among them and control group was statistically significant (P<0.001),the protein expression levels of Cyclin D1 and MMP-9 were increased,the protein expression level of E-cadherin was decreased.Conclusions Gal-3 is highly expressed in EOC and promotes the proliferation,migration and migration capability of EOC cell.
