龟鹿二仙胶对铅鼠海马CA_1区长时程增强损伤的修复作用

摘　　要：目的探讨龟鹿二仙胶对发育期铅暴露大鼠海马CA1区突触可塑性的影响。方法在出生当天将仔鼠随机分为正常组、正常+龟鹿二仙胶组、铅组及铅+龟鹿二仙胶组。仔鼠出生后饮用0.2%的醋酸铅溶液,建立铅鼠模型,采用石墨炉原子吸收光谱法测血铅,用液态等离子质谱法测定海马铅水平,在位电生理记录海马CA1区的场电位。结果和正常组相比,铅鼠血铅、海马铅水平明显升高,长时程增强(LTP)的幅度显著下降,均有显著性差异(P<0.01);而同期铅+龟鹿二仙胶组海马铅水平比铅组显著下降(P<0.01),其LTP幅度明显高于铅组大鼠(P<0.05)。结论龟鹿二仙胶对发育期慢性铅暴露大鼠LTP幅度的下降有修复作用。AIM To explore the effect of Guilu Erxian Gel on the lead-induced impairment of synaptic plastici- ty in hippocampal CA1 region of the developmental rats in vivo. METHODS On parturition day, neonatal rats were randomly divided into four experimental groups which included the control group, control with Guilu Erxian Gel, lead-exposed group, and lead-exposed with Guilu Erxian Gel group. The lead-exposed rats were with ad libi- turn access to 0. 2% lead acetate solution in the early postnatal period. The blood lead levels were analyzed by a PerkinElmer atomic absorption spectrometer and lead concentrations in hippoeampus were measured by a plas- maQuad plasma mass spectrograph. Field potential recordings in hippocampal CA1 region were performed in vivo. RESULTS Lead exposure caused an increase in blood and hippocampal lead levels accompanied by a dramatic decrease of long-term potentiation （LTP） amplitude compared to the control group （P 〈 0. 01 ）. After application of Guilu Erxian Gel, the hippoeampal lead concentrations were significantly lower than those of the lead group （P 〈 0.01 ）. Meanwhile, the treatment with Guilu Erxian Gel rescued lead exposure-induced impairment of LTP ampli- tude （P 〈 0. 05 ）. CONCLUSION Guilu Erxian Gel can rescue the impairment of LTP induced by chronic lead exposure.
