摘 要:目的观察槐定碱对骨癌痛大鼠脊髓N-甲基-D-天门冬氨酸(NMDA)受体亚型NR2B mRNA和神经元型一氧化氮合酶(nNOS)mRNA表达的影响,探讨其抗癌痛的作用机制。方法将W256癌细胞注入40只SD雌性大鼠骨髓腔复制骨癌痛模型后随机分为槐定碱治疗组(腹腔注射槐定碱25 mg/kg)及模型对照组,另取10只大鼠骨髓腔注入生理盐水,治疗前后分别测量大鼠热痛阈和机械痛阈;采用逆转录PCR(RT-PCR)检测大鼠脊髓NR2B mRNA和nNOS mRNA的表达。结果槐定碱能够明显提高W256癌细胞诱导的骨癌痛大鼠热痛阈和机械痛阈,明显下调骨癌痛大鼠脊髓组织NR2B mRNA和nNOS mRNA的表达。结论槐定碱抗骨癌痛作用可能与下调脊髓与痛觉过敏密切相关的NMDAR-nNOS/NO-cGMP信号转导通路有关。AIM To explore the effects of sophoridine on mRNA expression of N-methyl-D-aspartate (NMDA) receptor NR2B subunit and neuronal nitric oxide synthase (nNOS) in spinal cord of rats with bone cancer pain in- duced by W256 tumor cells, and their analgesic mechanism. METHODS Forty female Sprague-Dawley rats were used as models of bone cancer pain after being injected with W256 tumor cells into the marrow cavity, ran- domly divided into two groups: sophoridine treated group (25 mg/kg sophoridine) and model control group. Anoth- er ten rats were selected to be normal control group by injecting normal saline. Before and after the treatment, pain thresholds of thermal stimulation and mechanical stimulation were measured. After the treatment, the expression of NR2B mRNA and nNOS mRNA in spinal cord were detected by RT-PCR. RESULTS Sophoridine could obviously increase the thresholds of thermal stimulation and mechanical stimulation in rats with bone cancer pain induced by W256 tumor cells. And it could significantly downregulate the expression of NR2B mRNA and nNOS mRNA in spinal cord of rats with bone cancer pain. CONCLUSION The analgesic mechanism of sophoridine against bone cancer pain may be related to its downregulation of the signal transduction pathway of "NMDAR-nNOS/NO-cGMP" which is closely connected with hyperalgesia.
