摘 要:目的建立大鼠血清、肝脏、肾脏中补骨脂素、异补骨脂素定量测定的方法。方法采用乙酸乙醋-正己烷(8:1)为萃取溶剂进行样品处理,采用HPLC法测定,色谱柱AgilentTC—C18(2)(4.6mm×250mm,5μm);以氯霉素为内标物,流动相为水-乙腈,梯度洗脱,体积流量1mL/min,柱温30℃,检测波长295nm(0—10min,氯霉素),246nm(10~25min,补骨脂素、异补骨脂素)。结果补骨脂素、异补骨脂素在0.1~10.0mg/L范围内,在血清、肝匀浆液、肾匀浆液中均能达到很好的分离。结论本方法简便准确,可同时测定给药后大鼠血清、肝脏和肾脏中补骨脂素和异补骨脂素。AIM To establish a quantitative HPLC method for determining the contents of psoralen and isopso- ralen in rat' s serum, liver and kidney. METHODS The samples were extracted by ethyl acetate and n-butanol (8 : 1 ). The HPLC method was performed on the Agilent TC-C18 (2) and the mobile phase was water-aeetonitrile using chloramphenicol as the internal standard in the flow rate of 1.0 mL/min. The detection wavelength was set at 295 nm ( 0-10 min) for chloramphenieol, 246 nm (10-25 min) for psoralen and isopsoralen, the column temper- ature was 30 ℃. RESULTS The two components showed good linear relationship in the range of 0. 1-10. 0 mg/ mL, and could be separated in serum, liver and kidney of rats. CONCLUSION This method can be used to sim- ultaneously determine the content of psoralen and isopsoralen in serum, liver and kidney of rats.
