摘 要:目的:优选羌活-独活药对的合提工艺。方法:在单因素试验基础上,以总香豆素、蛇床子素和异欧前胡素提取量的综合评分为指标,通过正交试验考察乙醇体积分数、提取次数、料液比、提取时间对羌活-独活混合提取工艺的影响。采用UV测定总香豆素含量,检测波长315 nm;利用HPLC测定蛇床子素和异欧前胡素含量,流动相乙腈-水(45∶55),检测波长316 nm。结果:乙醇体积分数对合提工艺具有显著性影响,最佳提取工艺为加10倍量75%乙醇提取2次,每次1 h;总香豆素、蛇床子素、异欧前胡素提取量分别为470,24.46,9.73 mg·g-1。结论:优选的提取工艺稳定可行,为该药对的临床复方制剂开发提供参考。Objective : To optimize extraction technology for codecoction of Notopterygii Rhizoma et Radix coupled with Angelicae Pubescentis Radix. Method: Based on single factor tests, with composite score of extract amounts of total coumarins, osthole and isoimperatorin as index, orthogonal test was adopted to investigate effects of ethanol concentration, solid-liquid ratio, extraction times and time on extraction process. UV was used to determine the content of total coumarins with detection wavelength at 315 nm. Contents of osthole and isoimperatorin were measured by HPLC, mobile phase of acetonitrile-water (45 : 55 ), detection wavelength was set at 316 nm. Result: Ethanol concentration had a significant impact on extraction technology for codecoction, optimum conditions were as follows: extracted twice with 10 times the amount of 75% ethanol for 1 h per time; extract amounts of total coumarins, osthole and isoimperatorin were 470, 24.46 and 9.73 mg ·g-1, respectively. Conclusion: This optimized extraction process was stable and feasible, it could provide a reference for development of clinical compound preparation of this couplet medicines.
