摘 要:目的:研究盆康舒颗粒对大鼠慢性盆腔炎的保护作用及机制。方法:采用混合菌液注入大鼠子宫内,建立慢性盆腔炎动物模型,造模后大鼠随机分为5组,盆康舒(11.72,5.86,2.93 g·kg-1)组、模型组、妇炎康(2.25 g·kg-1)组,灌胃给药28 d后,测定WBC计数、子宫卵巢脏体比。采用酶联免疫吸附(ELISA)法测定血清TNF-a、IL-2含量。采用免疫组化法观察子宫内膜细胞Caspase-3、MMP-2、NF-kB表达。采用血流变仪测定全血血液流变学。子宫病理学检查。结果:与模型组比较,盆康舒颗粒各剂量组的TNF-a含量降低、IL-2含量升高(P<0.05);与模型组比较,高、中剂量组的WBC计数降低、Caspase-3、MMP-2的表达增加、全血血液黏度降低(P<0.01或P<0.05);各剂量组的NF-kB的表达和子宫卵巢脏体比虽有降低,但无统计学差异(P>0.05);各剂量组的子宫病理改变程度减轻。结论:盆康舒颗粒通过抗炎,改善血液循环,实现治疗目的。OBJECTIVE To study the anti-chronic pelvic inflammatory effect of Penkangshu granule and its mechanism of ac- tion in the rat chronic pelvic inflammatory disease (CPID) model, METHODS The CPID model was established by injection of mixed bacteria into two sides of the rat uterus. Then the rats were randomly divided into 5 groups as follows: Penkangshu granule low, middle and high groups (11.72,5.86,2. 93 g.kg-1^ ), model group, Fuyankang (2. 25 g.kg -1^ ) group. All the groups were continuously administrated for 28 days, the uterus ovary/body ratio, the content of TNF-a, IL-2 in blood serum, the expression level of Caspase-3, MMP-2, NF-kB in uterus pathological tissues, the parameters of hemorheology, the patholo- gy of uterus were determined. RESULTS Compared with model group, the content of IL-2 increased and TNF-a decreased sig- nificantly in Penkangshu granule each dose group(P〈0. 05). Compared with model group, WBC, whole blood viscosity de- creased and the level of expression of Caspase-3, MMP-2 increased in the high, middle-dose groups. Compared with model group, the ratio of uterus ovary/body and the level of expression of NF-kB had the trends of decreasing (P〉0. 05). The path- ological changes of uterus mentioned above were ameliorated in various degrees in all three dosage groups. CONCLUSION The objective of treatment of Penkangshu granule on chronic pelvic inflammatory disease was achieved by ameliorating inflammatory reaction and promoting blood circulation.
