摘 要:目的研究不同剂量刀豆蛋白A(ConA)对小鼠肝脏损伤的影响。方法60只小鼠被随机分为6组,对照组和试验组(3、6、9、12、15mg/kgBW)。对照组尾静脉注射医用生理盐水,试验组尾静脉注射剂量为3—15mg/kgBW的ConA,禁食8h后眼球采血,并取肝组织,进行各项指标的检测及病理组织学检查。结果随着ConA剂量的提高,小鼠肝脾肿大较为明显,肝脾指数与对照组相比,差异有统计学意义(P〈0.01)。血浆谷草转氨酶(AST)和谷丙转氨酶(ALT)在较高剂量下升高比较明显(P〈0.01);ConA剂量的增加对血浆还原性谷胱甘肽(GSH)的影响较为明显(P〈0.01),而对超氧化物歧化酶(SOD)影响并不是很大。肝匀浆中的糖原含量、GSH—PX以及SOD活性均有不同程度的降低,但与ConA剂量没有对应关系。肝脏病变在上述剂量范围内不明显。结论较高剂量的ConA可引起小鼠血浆和肝脏生化指标的显著变化,对肝脏病变影响不大。Objective The aim of the study was to investigate the effect of different doses of concanavalin A (ConA) on hepatic injuries in mice. Methods 60 mice were randomly divided into 6 groups, which included a control group and five experimental groups (3, 6, 9, 12, 15 mg/kg of BW). The control group was tail vein-injected with saline, while the experimental group was injected with 3-15 mg/kg BW of ConA. After 8 hours of fasting, blood was taken from eyes of the mice and hepatic tissues were removed for ex- amination of all biochemical indicators and histopathological examinations. Results With the increase in ConA dosage, splenohep- atomegalia of the experimental mice became more severe, and hepatosplenic indicators of the mice became significantly different from those of the control mice (P〈0.01). Plasma AST and ALT showed significant increase at higher ConA dosages (P〈0.01), and the increase in ConA strongly impacted the level of reduced glutathione (GSH) (P〈0.01) but not superoxide dismutase (SOD). In liver homogenates, glucogen and GSH-PX levels and SOD activity exhibited different degrees of reduction, which did not show to correlate with the dosage of ConA. Liver lesions were not significant under the above-mentioned dosage range. Conclusion High dosages of ConA can cause significant changes in biochemical indicators in plasma and liver of mice, but has little effect on lesions of the liver.
