MMPs、TNF-α、PLGF在子痫前期发病中的作用

摘　　要：目的:探讨子痫前期患者胎盘组织中基质金属蛋白酶(MMPs)、肿瘤坏死因子-α(TNF-α)、胎盘生长因子(PLGF)表达情况,分析其在发病中的作用。方法:选取我院50例子痫前期孕妇为研究组,并选取同期50例正常妊娠孕妇为对照组,检测两组胎盘组织中的MMPs、TNF-α、PLGF表达情况。结果:MMPs阳性显示:颗粒为棕黄色,主要分布在滋养细胞胞浆中,部分存在绒毛间质中;TNF-α阳性显示:颗粒为棕黄色,主要分布在滋养细胞胞浆与胞膜表面上;PLGF阳性显示:颗粒为棕黄色,主要分布在滋养细胞胞浆中。研究组中MMPs含量、PLGF含量明显的低于对照组,差异有统计学意义(P<0.05);研究组中TNF-α含量明显高于对照组,差异有统计学意义(P<0.05);轻度子痫前期和重度子痫前期的MMPs、TNF-α、PLGF含量比较,差异无统计学意义(P>0.05)。结论:子痫前期中TNF-α表达上调,MMPs和PLGF表达下调,三者在子痫前期发病中可能伴有协同的作用。Objective. To investigate expression of preeclampsia placenta tissue MMPs, TNF-α, PLGF in patients with preeclampsia and its role in the pathogenesis. Methods： A total of 50 pregnant women with preeclampsia admitted to our hospital were enrolled as study group and 50 healthy pregnant women were selected as control group. MMPs, TNF-α, PLGF expression in placental tissues were detec- ted. Results： MMPs positive was displayed as brown particles, mainly distributed in the cytoplasm of trophoblast cells, and some also presented in villous stoma. TNF-α -positive was presented as brown parti- cles, mainly distributed in the cytoplasm and membrane surface. PLGF positive was displayed as brown particles, mainly distributed in the cytoplasm of the trophoblast cells. MMPs and PLGF levels in the study group were significantly lower than the control group, the difference was statistically significant （P〈0.05） ;TNF- α level of the study group was significantly higher than that of the control group （P〈0.05）. MMPs, TNF-α, PLGF levels in mild preeclampsia and severe preeclampsia were not significantly different from each other （P〉0.05）. Conclusion： In preeclampsia patients, expressions of TNF-α was unregulated, but expressions of MMPs and PLGF were down-regulated, the three factors may cooperate with each other in the pathogenesis of preeclampsia.
