摘  要:目的探讨SH2-Bβ抗体对肥胖哮喘模型小鼠的治疗作用。方法采用8周C57BL/6小鼠(36只),随机分为正常组(标准饲料喂养)、肥胖哮喘组(高脂饲料喂养)和SH2-Bβ抗体干预组。测量各组小鼠体质量;应用免疫组织化学方法检测SH2-Bβ在各组小鼠肺内及下丘脑的表达;应用小鼠肺功能仪检测各组小鼠气道阻力变化。结果肥胖哮喘组体质量明显高于正常组和SH2-Bβ抗体干预组(P<0.05);免疫组织化学染色显示,在下丘脑,SH2-Bβ在肥胖哮喘组小鼠表达降低,明显低于SH2-Bβ抗体干预组和正常组(P<0.05)。在肺内,SH2-Bβ在肥胖哮喘组小鼠表达增高,明显高于SH2-Bβ抗体干预组和正常组(P<0.05);肥胖哮喘组小鼠的气道阻力明显高于SH2-Bβ抗体干预组和正常组(P<0.05)。结论 SH2-Bβ抗体可使肥胖哮喘模型小鼠下丘脑内SH2-Bβ表达上调,同时使肺内SH2-Bβ表达下调,降低体质量,降低肺内活化炎性细胞数量,进而降低气道阻力,使肥胖哮喘得到缓解。Objective To explore the therapy effects of SH2-Bβ antibody in the obese asthma mice. Methods Totally 36 C57BL/6 mice(female) aged 8 weeks were used throughout the study. Mice were randomized to 3 groups, including a standard rodent chow (NS, n =12) , a high fat diet chow obese asthma group ( OB+AS, n = 12) and the anti-SH2-Bβ intervene group. The weight was measured, and the SH2-Bβ expression in hypothalamus and lung was determined by immunohistorical staining. The changes of the airway resistance were evaluated by AniRes 2003 animal lungs function analysis system. Results The weight of OB+AS group was higher than that of NC group and anti-SH2-Bβ intervene group (P 〈 0.05 ). Immunohistochemistry staining demonstrated that SH2-Bβ expression in hypothalamus of OB + AS group were decreased compared with those of NC group and anti-SH2-Bβ intervene group (P 〈 0.05 ). SH2-Bβ expression in lung of OB+AS group were increased compared with those of NC group and anti-SH2-Bβ intervene group (P 〈 0.05 ). Airway resistance to methacholine (MCh) was significantly increased in OB+AS group compared with anti-SH2-Bβ intervene group (P 〈 0.05 ). Conclusion SH2-Bβ antibody upregulated the SH2-Bβ expression in hypothalamus, but downregalate its expression in lung.The obese asthma may be remitted through the decreased weight, number of the activated inflammatory cells and also airway resistance.
