摘 要:目的探讨展神经损伤后展神经核内神经再生的情况。方法选用健康成年Beagle犬22只,分为对照组(2只)和实验组。实验组又依据展神经损伤后1 d、3 d、7 d、14d、28 d分为五组(各4只),对照组和实验组经颞底入路显微手术,解剖、暴露展神经脑池段,仅实验组行展神经切断,术后测定展神经功能。对照组和实验组术后每条犬静脉注射BrdU 50 mg·kg-1,每天2次(早7∶00和晚7∶00),连续7 d。1 d组术后1 d给予注射BrdU,3 d组术后连续3 d注射BrdU,其他以此类推。术后1 d、3 d、7 d、14 d、28 d分别灌注取材,行nestin/BrdU、DCX/BrdU、GFAP/BrdU免疫荧光双标检测。结果实验组Beagle犬眼球呈内斜视,损伤侧展神经核内可见nestin/BrdU阳性神经前体细胞,DCX/BrdU阳性新生神经元前体细胞以及GFAP/BrdU阳性星形胶质细胞,BrdU阳性细胞术后3 d为高峰,术后7 d后逐渐下降,术后28 d时仍可以找到再生的神经细胞。对照组及实验组健侧展神经核内未见相关阳性细胞。结论展神经损伤后展神经核内出现神经再生现象。Objective To explore the neural regeneration in the abducens nucleus after abducens nerve injury. Methods Twenty-two healthy adult beagle dogs were selected and divided into control group (two dogs)and experimental group. The experimental group was sub-divided into 1 day group, 3 days groups, 7 days group, 14 days group and 28 days group after lesion, and four dogs in each group. All of the beagle dogs in two groups were anatomized and showed the cisternal segment of the abduceus nerve with microsurglcal subtemporal approach. Abducens nerves were cut only in experimental group and function test was measured. After operation, control group and experimental group were injected with BrdU(50 mg · kg^-1 ) ,2 times per day(7 : 00 am and 7 : 00 pm) for 7 days. The 1 day group and 3 days group received consecutive BrdU injection for 1 day and 3 days,respectively. Dogs were sacrificed after 1 day,3 days,7 days, 14 days,28 days,and nestin/BrdU,DCX/BrdU, GFAP/BrdU were examined by immunofluorescence. Results Ocular motility disorders caused by abducens nerves injury emerged in the injured eyes of beagle dogs. Nestin/BrdU double positive neural progenitor cells, DCX/BrdU double positive newborn neuronal progenitor cells and GFAP/BrdU double positive newborn astrocytes were only found in the abducens nucleus after abducens nerve injury. A peak of BrdU positive cell was found at 3 days and a decrease at 7 days. Most of the newly generated cells survived up to 28 days. There was no related positive cell in control group and uninjured side of experimental group. Conelusion Neural regeneration appears in the abducens nucleus of beagle dogs after abduceus nerve iniury.
