摘 要:目的:探讨人参皂苷Rg1对高糖诱导心肌细胞肥大的保护作用。方法:利用体外培养模型,以25 mmol/L葡萄糖诱导心肌细胞肥大,观察不同剂量人参皂苷Rg1(15、30、60 mol/L)对高糖诱导的心肌肥大的抑制作用。倒置荧光显微镜下观察细胞形态大小及肌丝改变,用考马斯亮蓝法测心肌细胞蛋白含量;用消化分离法及计算机图像分析系统测细胞体积;MTT法测细胞存活率;采用Till阳离子测定系统,以Fura-2/AM为荧光探针,观察胞内[Ca2+]i瞬间变化。结果:与正常对照组相比,高糖组使心肌细胞总蛋白质含量增加,细胞体积增大,细胞存活率降低,胞内[Ca2+]i瞬间变化的峰值增大。与模型组相比,人参皂苷Rg1(30、60mol/L)可有效改善高糖诱导的心肌细胞肥大,使心肌细胞存活率增高,同时抑制高糖诱导的细胞内[Ca2+]i瞬间变化的峰值,且其对胞内Ca2+的抑制作用与L-型钙通道抑制剂维拉帕米相似。结论:人参皂苷Rg1对高糖诱导的心肌细胞肥大有一定的保护作用,其作用可能与抑制细胞内Ca2+有关。Objective: The aim of the present study was to study the protective effect of Ginsenoside Rgl on high glucose induced myocardial hypertrophy. Methods: Myocardial ceils of neonatal rats were cultured in vitro. The hypertrophic myoeytes were induced by 25 mmol/L glucose. Then the effects of different concentrations of Ginsenoside Rgl ( 15,30,60 mol/L) on hypertrophic eardiomyoeytes were observed. Morphologic changes of cardiomyocytes were observed with inverted lluorescenee microscope; Total protein content was measured by the method of Brad- ford. The cardiomyoeytes size was measured by computer photograph analysis system. The cardiaomyoeytes viability was analyled by MTF; [ Ca2 + ]i transient was measured by Till image system by cell-loading Fura-2/AM. Resdts: Compared with control group, high glucose in- creased the total protein content, the cardiomyoeytes size, [ Ca2+ ]i transient, and decreased the cell viability of cardiomyoeytes. Compared with the model group, The total protein content, cell size were decreased, and the cell viability was increased by Ginsenoside Rgl (30、60 mol/L). In addition the increased [ Ca2+ ]i transient was decreased by Ginsenoside Rgl (30、60 mol/L) ,which has similar effect with verapamil. Conclusion: Ginsenoside Rg1 has a protective effect on high glucose induced myocardial hypertrophy, which is partially via regulating intracellular Ca2+.
