摘 要:目的:观察绿原酸(chlorogenic acid)对血管生成的抑制作用及其机制。方法:采用鸡胚尿囊膜(CAM)模型观察绿原酸对血管生成的抑制作用。采用MTT、Transwell及Western blotting方法研究绿原酸抑制血管生成的机制。结果:100μg/ml绿原酸能明显抑制CAM新生血管的生长。绿原酸能抑制HUVEC细胞增长的IC50为38μmol/L;绿原酸浓度为25μg/ml、50μg/ml、100μg/ml时,HUVEC细胞迁移数和VEGF蛋白表达量均受到抑制,且呈剂量依赖性。结论:绿原酸能够抑制血管生成,该作用可能与下调血管内皮生长因子VEGF的表达有关。Objective: To observe the anti-angiogenesis effect of chlorogenic acid and discuss the possible mechanism. Method: Chick embryo chorioallantoic membrane( CAM) model was used to detect the anti-angiogenetic effect of chlorogenic acid in vivo. MTT,Transwell and Western blot were used to detect the mechanism in vitro. MTT was used to measure the proliferation of HUVEC. Transwell was used to detect the migration of HUVEC. The expression of VEGF was determined by Western blotting. Result: Cholrogenic acid at dose of 100 μg /ml had significant inhibitory effect on CAM angiogenesis. The proliferation of HUVEC was inhibited by cholrogenic acid in a dose-dependent manner( IC50= 38μmol /L). When the concentrations of chlorogenic acid were 25 μg /ml,50 μg /ml,100 μg /ml,the migration of HUVEC and the expression of VEGF were inhibited in a dose-dependent manner. Conclusion: Chlorogenic acid had anti-angiogenetic effect in vivo and in vitro,and the action mechanism may be associated with decreasing the expression of VEGF.
