青钱柳叶总黄酮对自发性高血压大鼠的影响

摘　　要：目的:探讨青钱柳叶(Cyclocarya paliurus)总黄酮对自发性高血压大鼠(SHR)的降压作用。方法:采用无创尾动脉测压法观察青钱柳叶总黄酮对SHR收缩压(SBP)、舒张压(DBP)、心率(HR)、体重、脏器指数的影响,测定血常规及血清超氧化物歧化酶(T-SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮(NO)、一氧化氮合酶(TNOS)、心房钠尿肽(ANP)含量,观察SHR肾脏HE染色病理变化、肾小球电镜检查、Ki-67、VEGF在肾脏中的免疫组化表达,左心室、主动脉和肾主动脉组织形态学变化。结果:青钱柳叶总黄酮低、中、高剂量组(3.0,6.0,12.0 g/kg)能明显降低SHR的SBP和DBP,并对HR无明显影响;能够缓解左心室、肾脏、脾脏和主动脉病变,提高RBC、HGB、GSH-Px、NO、TNOS含量;降低血浆ANP含量。对肾脏中Ki-67、VEGF的阳性表达有明显抑制作用,在某种程度上具有逆转SHR靶器官损害的作用。结论:青钱柳总黄酮具有一定的降血压的作用,其机制可能与升NO、NOS,ANP。Objective： To study the effect of the flavonoids from Cyclocarya paliurus on spontaneous hypertension rats. Method： Non-invasive blood pressure measurement was used to detect the SBP,DBP and HR. Then the body weight,organ index,blood,the T-SOD,MDA,GSH-Px,NO,TNOS and ANP content were measured. Observe SHR kidney HE staining pathological changes,electron microscopic examination of glomeruli,immunohistochemical expression of Ki-67 and VEGF in the kidney,histological changes of the left ventricle,aorta and renal artery. Result：The low,medium and high dose group（ Dose were： 3. 0,6. 0,12. 0g /kg） of flavonoids from Cyclocarya paliurus can significantly lower SBP and DBP in SHR,but had no obvious effect on HR. It can alleviate the lesions of the left ventricle,kidneys,spleen and aorta,improve RBC,HGB,GSH-Px,NO and TNOS content,and lower plasma ANP levels. The positive expression of Ki-67 and VEGF in kidney was significantly inhibited,to some extent it had the role to reverse the SHR target organ damage. Conclusion： The flavonoids from Cyclocarya paliurus could reduce blood pressure of SHR. The mechanism may be associated with the elevation of the NO,NOS and reduction of the ANP content.
