摘 要:目的:探讨丹参水提物(SME)对大鼠缺血-再灌注损伤后氧化应激的影响。方法:将50只大鼠随机分为5组:假手术组、缺血-再灌注(MI/R)模型组、1.21g/kg复方丹参片组、0.02976 g/kg及0.05952 g/kg丹参水提物组。左冠状动脉前降支结扎30min后实现再灌注2h采血,测定血清乳酸脱氢酶(LDH)、肌酸激酶(CK)、谷草转氨酶(GOT)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、活力。取心脏组织,剪去心房及右心室,TTC染色,计算梗死范围。结果:与假手术组相比,缺血-再灌注损伤组血清SOD、GSH-Px活力降低,MDA、LDH、CK、GOT活力升高;心肌组织梗死面积升高。与缺血-再灌注损伤组相比,0.02976 g/kg及0.05952 g/kg丹参水提物组和1.21 g/kg复方丹参片组血清SOD、GSH-Px活力升高,MDA、LDH、CK、GOT活力降低,心肌组织梗死面积降低。与复方丹参片组相比,0.05952 g/kg丹参水提物组可以明显改善上述各项指标。结论:丹参水提物可通过改善机体抗氧化能力对大鼠缺血-再灌注损伤具有保护作用。Objective: To study the effects of Salvia miltiorrhiza extract( SME) on oxidative stress after myocardial ischemia-reperfusion injury in rats. Methods: Fifty rats were divided into five groups: control group,MI / R,treatment with SME( 0. 02976 g / kg and 0. 05952 g / kg by gavage),and treatment with FDT( 1. 21g / kg by gavage),MI / R was produced by occluding the left coronary artery for 30 min followed by reopening the artery for 120 min. At day 3,serum was stored at-80℃ for further cardiac enzymes and parameters of oxidative stress were measured. The myocardial infarct size was measured using triphenyltetrazolium chloride( TTC) staining. Results: MI / R injuried rats showed increased in serum levels of LDH、GOT,CK and MDA,as well as decreased in serum activities of GSH-Px and SOD( P < 0. 05,P < 0. 01). Oral administration of SME( 0. 02976 and 0. 05952 g / kg) blunted all of the biochemical changes induced by MI / R,as did FDT( 1. 21g / kg)( P <0. 05,P <0. 01). The protective effect of SME on MI/R-induced myocardial damage was further confirmed by myocardial infarct size examination. And SME 0. 05952g / kg doing so to a greater degree than FDT( P < 0. 05,P < 0. 01). Conclusion: SME affords protection against MI / R-induced myocardial infarction in rats.
