摘 要:目的:观察姜黄素对慢性阻塞性肺病患者外周血内皮祖细胞一氧化氮分泌和内皮型一氧化氮合酶表达的影响,探讨姜黄素对慢性阻塞性肺病患者血管保护作用的机制。方法:选取住院慢性阻塞性肺病患者30例,诱导外周血单个核细胞分化为内皮祖细胞,消化收集贴壁细胞,加入不同浓度姜黄素(分别为5,10,15μmol/L),并设立不加入姜黄素的对照组,相同条件下培养一定时间(6小时,12小时,24小时和48小时)。鉴定荧光双染色阳性的细胞为内皮祖细胞,使用Griess法检测一氧化氮分泌量,Western Blot法检测一氧化氮合酶蛋白的表达。结果:姜黄素显著改善了慢性阻塞性肺病患者内皮祖细胞的一氧化氮分泌功能,呈浓度时间依耐性,当姜黄素浓度为15μmol/L,共培养24h后改善最为明显。姜黄素也显著增强了慢性阻塞性肺病患者内皮祖细胞的一氧化氮合酶表达,姜黄素浓度为15μmol/L时最为显著。结论:姜黄素促进了内皮祖细胞的一氧化氮分泌量并增强了一氧化氮合酶的表达,提示姜黄素可通过改善内皮祖细胞的功能活性来修复慢性阻塞性肺病患者损伤的血管内皮功能。Obiective: To investigate whether curcumin has influence on on NO secretion and eNOS expression in endothelial progenitor cells( EPCs) of COPD patients. Methods: The peripheral blood mononuclear cells from peripheral blood of 30 COPD patients were induced into EPCs and stimulated with curcumin( to make a series of concentrations: 0μmol /L,5μmol /L,10μmol /L,15μmol /L) for the respective time points( 6h,12h,24h,48h). EPCs were characterized as adherent cells double positive by fluorescent staining. Nitric oxide secretion were assayed with Griess Reagent System. Western Blot was used to measure the expression of endothelial nitric oxide synthase. Results: Curcumin significantly improved the NO secretion in endothelial progenitor cells of COPD patients within concentration and time dependence,maximum at 15μmol /L,24h( P < 0. 01). The expression of eNOS was also be significantly up-regulated,maximum at 15μmol /L as well( P < 0. 01).Conclusion: Curcumin enhances the NO secretion and eNOS expression in EPCs,suggesting that curcumin could repair the injured vascular endothelial function of COPD patients by promote the function and activity of EPCs.
