摘  要:为了解鸡鼻相关淋巴组织(NALT)不同时期的免疫状态,本实验选择不同日龄的海兰白鸡的鼻组织,利用免疫组织化学方法研究CD3+T淋巴细胞和Bu-1+B淋巴细胞的出现、定位分布及数量变化过程。结果显示,Bu-1+细胞和CD3+细胞最初均于胚胎20 d出现。在定位分布上,NALT以鼻泪管开口处最为发达,4日龄时NALT出现在鼻泪管周围,CD3+T淋巴细胞便成为NALT中的主要淋巴细胞群,并且在14日龄以后,T、B淋巴细胞的分布更具区域化,即CD3+T淋巴细胞聚集在鼻黏膜上皮下固有层底端,占据NALT的中下部区域,而Bu-1+细胞则占据中上部区域。在数量变化上,CD3+T淋巴细胞数量在14日龄以前各日龄呈逐渐增加趋势,而从21日龄开始又逐渐减少;Bu-1+B淋巴细胞则整体呈逐渐增加趋势。而且14日龄之前CD3+细胞数量多于Bu-1+细胞,而此后各日龄Bu-1+细胞的数量均显著多于CD3+细胞。结果表明,鸡NALT中T、B淋巴细胞的分布和数量均呈现日龄相关性变化,并且其变化可以反映出14日龄之前NALT以细胞免疫为主,而之后则倾向于体液免疫。To investigate the immune state of chicken nasal-associate lymphoid tissue (NALT) in different age, the location, distribution and quantity change of T and B lymphocytes was examined in the nose tissue of Hy-line Variety White chicken at different ages by immunohistochemistry staining method. The results showed that Bu-1^+ cells appeared initialy at 20 embryonic days,while CD^3+ cells were detectible at 1-day-old. In addition, NALT were found the most developed in the openings of nasolacrimal duct and the first appearance of NALT around the nasolacrimal duct and CD3^+ T cells formed the main lymphocyte population in NALT, and after 14-day-old, the distributions of T and B cells were more regionalization, that CD3^+ T cells gathered at the bottom of the lamina propria of the nasal mucosa and occupied in the middle and inferior part of NALT, and Bu-1+ cells distributed in the middle and superior part of NALT. The amount of CD3+ T cells increased gradually before 14-day-old, and then gradually decreased from 21-day-old; Bu-1^+ B cells increased gradually in the overall trend. And the number of CD3^+ cells were more than Bu-1+ cells before 14-day-old, and thereafter the number of Bu-1+ cells were significantly more than CD3+ cells. The results reveal that the distribution and amount of T and B cells in chicken NALT were age related changes, and the changes might reflect that the immunity of NALT are mainly cellular immunity, and then tend to humoral immunity.
