摘 要:目的研究岗梅根总皂苷的最佳提取工艺条件。方法采用Design Expert 8.0分析软件,在单因素的基础上运用Box-Behnken响应曲面设计法,以提取时间、乙醇浓度、溶剂用量和提取次数建立4因素3水平的实验模型,并确立提取回归方程;使用响应曲面法优化提取工艺。结果最佳的工艺条件为:提取时间100 min,乙醇体积分数为56.33%,溶媒用量为11倍,提取次数为2次。验证实验显示总皂苷平均转移率为71.92%,模型预测值的绝对误差小于3%。结论响应曲面法能合理地优化岗梅中总皂苷的提取工艺,可为岗梅的综合利用提供参考。Objective To optimize the extraction technology for total saponins from the roots of Ilex asprella. Methods On the basis of the single factor lest, the effects of extracting time, volume of ethanol, solid/liquid ratio and extraction times on the saponin process were studied by the orthogonal test developed by Box-Behnken central composite test design, and the regression equation was also established. The results were analyzed, and the extracting process was optimized by the response surface methodology( RSM ). Results The optimal conditions were as follows: extraction with 56 % of ethanol solution for 100 rain, solid/liquid ratio being 1 : 11 (m/v) and extracting times being twice. The results of validation test showed that the average yield was 71.92 %. The absolute error of model predicted value was less than 3 %. Conclusion RSM can be used for the optimization of extracting technology for total saponins from the roots of Ilex asprella, which will supply evidence for the multiple utilization of Ilex asprella.
