摘  要:目的 : 研究视黄酸 (RA)促进脐血淋巴细胞 (CBL )抗体生成的作用机制。方法 : 对脐血标本进行血清维生素 A(VA)浓度测定 ,采用 RT- PCR法检测 CBL中视黄酸受体 -α(RAR-α)的基因表达水平 ,并采用 ELISA法检测细胞培养上清液中 Ig M含量。结果 : 脐血血清 VA水平与未培养 CBL中 RAR- α的基因表达水平之间存在正相关 :r=0 .50 (P<0 .0 1 ) ;RA增加金黄色葡萄球菌 Cowan 株 (SAC)活化 CBL的 Ig M生成能力 (P<0 .0 0 1 ) ;RA上调 SAC活化 CBL中RAR- α的基因表达水平 (P<0 .0 0 1 ) ;RA使 RAR- α基因表达的上调幅度与 RA使 Ig M生成的上调幅度之间存在正相关 :r=0 .40 (P<0 .0 5)。结论 :  RA对Objective: To further explore the mechanism by which retinoic acid (RA) promotes the antibody production of cord blood lymphocytes (CBL). [WT5FZ]Methods: The levels of serum vitamin A in cord blood were measured. The expression of retinoic acid receptor α (RAR α) gene was determined by RT PCR (reverse transcription polymerase chain reaction). The IgM concentrations in the supernatants of cell culture were measured by ELISA. [WT5FZ]Results: A positive correlation was found between serum vitamin A level and RAR α expression level in uncultured CBL:r=0.50 (P<0.01). IgM synthesis of SAC activated CBL was augmented by RA (P<0.001). RAR α expression level of SAC activated CBL was increased by RA (P<0.001). RAR α expression level of SAC activated CBL was increased by RA (P<0.01). There was a positive correlation between the enhanced amplitude of RAR α expression and the enhanced amplitude of IgM synthesis by RA:r=0.40 (P<0.05). [WT5FZ]Conclusion: Our studies suggest that RAR α pathway perhaps is one of main ways by which RA promotes antibody production of CBL.
