摘 要:目的探讨蜘蛛香环烯醚萜类成分对肠易激综合征(IBS)模型大鼠5-羟色胺(5-HT)、色氨酸羟化酶1(TPH1)以及单胺氧化酶A(MAO-A)的影响。方法 SD雄性大鼠随机分为对照组、模型组、匹维溴铵组和蜘蛛香环烯醚萜类成分高、中、低剂量(0.60、0.30、0.15 mg/kg)组,采用慢性应激方法建立IBS大鼠模型,观察蜘蛛香环烯醚萜类成分对各组大鼠腹部回撤反射压力阈值的影响。采用高效液相-电化学法测定血清5-HT的含量,RT-PCR检测各组大鼠结肠TPH1mRNA的表达,比色法测定结肠MAO-A活性。结果模型组大鼠的疼痛压力阈值和最大耐受压力阈值显著降低(P<0.01),蜘蛛香环烯醚萜类成分各剂量均可显著增加模型大鼠压力阈值(P<0.01)。模型组大鼠血清5-HT水平升高(P<0.05),结肠TPH1mRNA表达水平升高(P<0.01),MAO-A活性降低(P<0.05)。蜘蛛香环烯醚萜类成分中剂量以及匹维溴铵均可显著降低血清5-HT的水平以及结肠TPH1mRNA表达,并显著增加MAO-A的活性(P<0.01,P<0.05);蜘蛛香环烯醚萜类成分低剂量可显著降低结肠TPH1mRNA表达(P<0.05),并显著增加MAO-A的活性(P<0.05)。结论蜘蛛香环烯醚萜类成分可通过对结肠TPH1和MAO-A的调节作用,影响血清5-HT表达水平,降低IBS大鼠内脏敏感性。Objective To investigate the influences of iridoid of Zhizhuxiang (Jatamans Valeriana Rhizome) on the changes of 5-hydroxytryptamine (5-HT), tryptophan hydroxylase 1 (TPH,) and monoamine oxidase-A (MAO-A) in rats with irritable bowel syndrome (IBS). Methods Male SD rats were randomly divided into control group, model group, pinaverium group, and high-dose (0.6 mg/ kg), mid-dose (0.3 mg/kg) and low-dose (0.15 mg/kg) iridoid groups (high-dose group, mid-dose group and low-dose group). The rat model of IBS was established by using chronic stress method. The influence of iridoid on the threshold of abdominal withdraw reflection (AWR) was observed in all groups. The content of serum 5-HT was detected by using HPLC-EC, the expression of colon TPH~ mRNA was detected by using RT-PCR, and activity of colon MAO-A was determined by using colorimetric method. Results The pain pressure threshold and maximum tolerance pressure threshold decreased significantly in model group ( P 〈 0.01 ), and increased significantly in 3 iridoid groups ( P 〈 0.01 ). In model group, the content of serum 5-HT increased ( P 〈 0.05 ), expression of colon TPH1 mRNA increased (P〈O. 01 ) and activity of MAO-A decreased (P 〈0.05). In mid-dose group and pinaverium group, the content of serum 5-HT and expression of colon TPH1 mRNA decreased significantly and activity of MAO-A increased significantly ( P 〈 0.01 or P 〈 0.05 ). In low-dose group, the expression of colon TPH~ mRNA decreased significantly ( P 〈 0.05 ) and activity of MAO-A increased significantly ( P 〈 0.05). Conclusion Iridoid of Zhizhuxiang can affect the content of serum 5-HT and reduce vislral sensitivity in rats with IBS through regulating colon TPH1 and MAO-A.
