柴胡渗湿汤对肺纤维化大鼠肺组织MMP-9及TIMP-1表达的影响

摘　　要：目的探讨柴胡渗湿汤治疗肺纤维化机制。方法 Wistar大鼠90只,随机分为正常对照组、模型对照组、强的松组、柴胡渗湿汤组、柴胡渗湿汤加强的松组。除正常对照组外,均采用经支气管注入博莱霉素法建立肺纤维化大鼠模型。动态对比各组大鼠第14天、28天肺组织MMP-9、TIMP-1蛋白的表达。结果与正常对照组比较,模型对照组第14天、28天MMP-9、TIMP-1阳性颗粒表达明显增多,差异有统计学意义(P<0.05)。与模型对照组比较,各治疗组大鼠MMP-9、TIMP-1阳性颗粒表达及MMP-9/TIMP-1比值均明显降低,差异均有统计学意义(P<0.05)。与强的松组比较,柴胡渗湿汤组大鼠MMP-9、TIMP-1阳性颗粒表达接近,差异无统计学意义(P>0.05);而柴胡渗湿汤加强的松组MMP-9、TIMP-1阳性颗粒表达降低,差异有统计学意义(P<0.05)。与柴胡渗湿汤组比较,柴胡渗湿汤加强的松组MMP-9、TIMP-1阳性颗粒表达降低,差异有统计学意义(P<0.05)。肺组织病理形态学改变亦说明各治疗组优于模型组。结论柴胡渗湿汤可能通过降低MMP-9、抑制TIMP-1表达,调节两者比例的平衡来治疗肺纤维化,与强的松联合后效果有一定程度加强。Objective To investigate the mechanism of ＂Chaihu Shenshi Decoction＂ for the treatment of pulmonary fibrosis. Methods Ninety Wistar rats were randomly divided into normal control group, model group, prednisone group, ＂Chaihu Shenshi Decoction＂ group, ＂Chaihu Shenshi Decoction＂ plus prednisone group. Except the normal control, the rest of groups were injected with bleomycin by trachea to set up the model of pulmonary fibrosis. We compared the expression of MMP-9 and TIMP-1 in lung tissues of rats at the 14th and 28th day. Results Compared with normal control group, the expression of MMP-9 and TIMP-1 was increased obviously al the 14th and 28th day in model control group, with significant difference between the two groups （ P 〈 0.05 ）. Compared with model group, the expression of MMP-9 and TIMP-1 and the ratio of MMP-9/TIMP-1 in treatment group were reduced, with significant differences between the two groups （ P 〈 0.05 ）. No significant difference was found in expression of MMP-9 and TIMP-1 between ＂Chaihu Shenshi Decoction＂ and prednisone groups （ P 〉 0.05 ） , and expressions of MMP-9 and TIMP-1 were reduced in ＂Chaihu Shenshi Decoction＂ plus prednisone group, was significant different from that of the prednisone group （ P 〈 0.05 ）. There were significant differences in expression of MMP-9 and TIMP-I between ＂Chaihu Shenshi Decoction＂ and＂ Chaihu Shenshi Decoction＂ plus prednisone group （ P 〈 0.05 ）. The morphological changes of lung tissues in treatment group were better than that of the model group. Conclusion Chaihu Shenshi Decoction＂ has therapeutic effects on pulmonary＂ fibrosis, which may be related to reducing MMP-9, inhibiting TIMP-1 expression and regulating the ratio of MMP-9 and TIMP-1. The therapeutic effects can be strengthened by combining with prednisone.
