麻杏石甘汤对3型腺病毒感染HELF部分细胞因子mRNA基因表达的影响

摘　　要：目的:探讨麻杏石甘汤含药血清对3型腺病毒感染的人胚肺成纤维细胞(HELF)之转化生长因子-β1(TGF-β1)、血小板衍生生长因子-BB(PDGF-BB)、肿瘤坏死因子-α(TNF-α)mRNA基因表达的影响。方法:以3型腺病毒分别攻击体外培养的HELF,制备麻杏石甘汤含药血清,作用于该细胞,另设正常细胞组、病毒对照组、利巴韦林组,采用原位杂交法检测不同组细胞TGF-β1、PDGF-BB、TNF-α的mRNA基因表达,进行各组间比较。结果:病毒对照组较正常细胞组之TGF-β1、PDGF-BB、TNF-αmRNA的表达均明显升高(P<0.01),含药血清组对腺病毒攻击后的TGF-β1、PDGF-BB、TNF-αmRNA表达均有显著降低作用(P<0.01)。结论:麻杏石甘汤可下调TGF-β1、PDGF-BB、TNF-α的mRNA表达,这可能是其治疗病毒性肺炎的作用机制之一。Objective ： To study the effect of Maxing Shigan Decoction contained serum on some cytokine mRNA Gene expressions of HELF induced by adenovirus type3. Method ： We determined the mRNA Gene expressions of transforming growth factor- β1 （ TGF- β1）, platelet-derived growth factor-BB （ PDGF-BB ） and tumor necrosis fsctor-α （ TNF-α ） in ADV-infected human embryonic lung fibroblast cells before and after adding the contained-herbs serum in situ hybridization. Results ： Adenovirus could up-regulate the protein expressions of TGF-β1, PDGF-BB and TNF-α in human embryonic lung fibroblast cells（ P〈0.01 ）. However, Maxing Shigan Decoction could decrease them （ P〈0.01 ）. Conclusions ： Maxing Shigan Decoction could regulate some cytokine mRNA levels. It might be a part of the mechanism of the therapy of Maxing Shigan Decoction in treating viral pneumonia.
