摘  要:为了探讨牛TRAF6基因的结构与功能,采用基因克隆技术分离了牛TRAF6基因的编码区序列,并采用生物信息学方法分析了该基因及其所编码的蛋白质的序列特征、结构和功能。结果表明:分离了TRAF6基因1803 bp的cDNA序列(GenBank登录号为DQ471669),编码区长度为1 629 bp,编码542个氨基酸(GenBank登录号为ABF06558),位于细胞核、细胞质、线粒体、内质网等多种细胞器上,分子量为61.57176 ku,等电点为6.09,含有RING-finger、zf-TRAF和MATH_TRAF6结构域。该蛋白与人、小鼠和大鼠TRAF6蛋白的同源性分别为89%、84%和82%,其结构域的同源性均达到95%以上。根据各物种间TRAF6蛋白的高同源性,结合人和小鼠TRAF6蛋白在TNF/TLRs/TIR超家族介导的信号转导途径中的重要作用,推测牛TRAF6蛋白也可能参与调节多种信号通路,在免疫反应、炎症反应、细胞分化和凋亡等生物学过程中发挥重要功能。In order to clarify the structure and function of bovine TRAF6 gene, the coding region sequence (CDS) of TRA F6 gene of diary cattle was isolated by gene cloning technology, and its character, structure and function were analyzed by bioinformaties methods. The results showed that 1 803 bp cDNA (GenBank ID: DQ471669) of coding sequence of cattle TRA F6 gene was isolated, which included the complete 1 629 bp CDS, coded 542 amino acids (GenBank ID: ABF06558), and located on many cell organs, including cell nucleus, cytoplasm, mitochondria and endoplasmic retieulum. The molecular weight and isoeleetric point of diary cattle TRAF6 protein were 61.57176 ku and 6.09, respectively. TRAF6 protein contained several domains including RING-finger, zf-TRAF and MATH_TRAF6. The homologies of TRAF6 protein between diary cattle and human, mouse, rats were 89%, 84% and 82%, respectively. The homologies of protein domain among all species mentioned above were up to 95%. According the results and the key functions of TRAF6 protein for many signal transduction pathways mediated by TNF/TLRs/TIR super families in human, it could infer that diary cattle TRAF6 might be an important signal molecular for regulating many signal transduction pathways, and might be served as very important functions for many biological processes, like immune response, inflammation, cell apoptosis and differentiation.
