摘 要:目的探讨水通道蛋白8(aquaporin 8,AQP8)和细胞外信号调节激酶磷酸化(extra-cellular regulated protein kinase phosphorylation,P-ERK1/2)在维吾尔族宫颈病变组织中的表达差异及其与宫颈癌临床病理参数的关系。方法应用免疫组织化学方法检测AQP8、ERK1/2、P-ERK1/2在108例子宫颈癌、47例宫颈上皮内瘤变(CIN)、79例正常宫颈组织中的表达,并分析其与临床病理参数的关系。结果 AQP8和EKR1/2的表达主要定位于细胞质内,P-ERK1/2的表达主要定位于细胞核内。AQP8在子宫颈癌、CIN及正常宫颈组织中的阳性表达率分别是46.30%、80.90%、15.20%;ERK1/2在3组间的阳性表达率分别是60.10%、76.60%、25.30%;P-ERK1/2在3组间的阳性表达率分别是44.40%、66%、24.10%;AQP8与宫颈癌的浸润深度有关,EKR1/2和P-ERK1/2与临床分期和远处转移有关;AQP8与ERK1/2在宫颈癌组织中的表达呈正相关。结论 AQP8、EKR1/2和P-ERK1/2蛋白表达上调可能与宫颈癌早期浸润和转移有关,AQP8与ERK1/2、P-ERK1/2可能成为宫颈癌早期诊断的候选标志物。[Objective] To probe differential expression of aquaporin 8 (aquaporin 8, AQP8) and extracellular regulated protein kinase phosphorylation (P-ERK1/2) in Uighur cervical lesions and its relationship with clinieopathological parameters in cervical cancer. [Methods] Immunohistoehemistry was used to detect expression of AQP8, ERK1/2 and P-ERK1/2 in 108 cases of cervical cancer, 47 cases of CIN and 79 cases of normal cervical tissues, and the relationship with elinieopathologieal parameters was analyzed. [Results] Expression of AQP8, EKR1/2 was mainly located in the cytoplasm, expression of P-ERK1/2 was mainly located in the nucleus. Expression rate of AQP8 in cervical cancer, CIN and normal cervical tissues was 46.30%, 80.90%, 15.20%, respectively; expression rate of ERK1/2 among three groups was 60.10%, 76.60%, 25.30%, respectively; expression rate of P-ERK1/2 among three groups was 44.40%, 66.00%, 24.10%, respectively; expression of AQP8 was related with invasive depth of cervical cancer, and expression of EKR1/2 and P-ERK1/2 was related with clinical stage and lymphatic metastasis; expression of AQP8 was positively correlated with ERKln in cervical cancer. [Conclusion] Up-regulated expression of AQP8, ERK1/2 and P-ERK1/2 may be associated with invasion and metastasis in early stage of cervical cancer, AQP8, ERK1/2 and P-ERK1/2 may become candidate markers for early diagnosis of cervical carcinoma.
