摘  要:目的:观察乌司他丁(ulinastatin,UTI)提前给药对过氧化氢(H2O2)诱导H9c2心肌细胞损伤的影响。方法:采用体外培养的H9c2心肌细胞进行实验,通过500μmol/L H2O2与细胞共孵育24 h建立心肌细胞氧化应激损伤模型。实验分为正常对照组(NC组)、氧化应激模型组(H组)、乌司他丁组(U组)和乌司他丁提前给药组(UH组)。实验处理后,吉姆萨染色倒置显微镜观察各组细胞形态差别,MTT法测定细胞活力,流式细胞技术检测细胞凋亡情况,测定各组细胞超氧化物歧化酶(SOD)和丙二醛(MDA),及检测培养液上清中乳酸脱氢酶(LDH)活力。结果:与NC组比较,H组细胞出现明显损伤,细胞存活率显著降低、细胞凋亡率升高,SOD活力降低,MDA含量增加,培养液上清中LDH活力明显升高(P<0.01)。而与H组比较,UH组细胞存活率升高,细胞凋亡率降低,SOD活力升高,MDA含量及LDH活力降低(P<0.01)。结论:乌司他丁提前给药对H2O2诱导性H9c2细胞氧化应激损伤有一定的保护作用。Objective To observe the effects of the preadministration of ulinastatin on H9c2 ceils after H2O2-induced oxidative stress. Methods H9c2 cells were cultured in vitro and divided into 4 groups: normal control group (Group NC), H2O2 oxidative stress group (Group H), ulinastatin group (Group U) and preadministration of ulinastatin group (Group UH). H2O2 oxidative stress injury models were established by co-culture of the H9c2 cells with H202 (500 μmol/L). The differences in morphology of the ceils were observed under the inverted microscope and the cell viability and apoptosis were determined by MTr and flow cytometry, respectively. The activity of superoxide dismutase (SOD) and the contents of malonadehyde (MDA) of the cells were determined by spectra photometric assay. Supernatant LDH activities in the supematant of H9c2 were determined. Results As compared with those in Group UH, the viablity of H9c2 and the activity of SOD and LDH were decreased (all P 〈 0.05, respectively), the apoptosis rate and the content of MDA were increased (both P 〈 0.05, respectively). Conclusion The preadministration of ulinastatin could effectively inhibit the oxidative stress injury of H9c2 cells induced by H2O2.
