摘  要:目的:探讨贴壁培养细胞直接用培养瓶置4℃、-20℃,最后置-80℃低温冰箱冻存的可行性。方法:采用培养瓶常规培养贴壁细胞L929,去上清后,加冻存液覆盖细胞。-80℃低温冰箱冻存,然后,每2个月复苏1次,计数活细胞,观察细胞传代效果。结果:L929细胞置-80℃低温冰箱冻存1年,复苏良好,随着冻存时间延长,存活细胞逐渐减少,复苏耗时延长。结论:贴壁培养细胞直接用培养瓶置-80℃低温冰箱冻存1年,复苏良好。Objective: To explore the feasibility of adherent cells to preserve at -80 ℃ refrigerator directly with flask. Methods: The adherent cell line L929 was normally cultured in the flasks, then the supernatant was deleted and cell freezing medium was added to cover the cells. The flasks were placed at 4 ℃, -20 ℃, -80 ℃ refrigerators, then recovered 1 in every 2 months and counted the living cells and observed the effect of cell passage. Results: L929 cells had been stored at -80 ℃ for one year and recovered successfully. With the freezing time prolonged, the living cells decreased, the recovery time increased. Conclusion: Adherent cells could be preserved at -80 ℃ refrigerator directly with flask for 1 year, and their recovery effect is good.
