急性心肌梗死合并室颤与MMP-9相关性探讨

摘　　要：目的探讨急性心肌梗死合并室颤与MMP-9表达的相关性。方法选择我院10例急性心肌梗死合并室颤患者纳入实验组;同期选择本院10例急性心肌梗死未合并室颤患者作为对照组。实验组及对照组于发病后第1、3、5d各抽取外周静脉血5ml,-20℃冰箱保存。采用RT-PCR及ELISA法同时测定MMP-9。结果 RT-PCR结果显示实验组第1d血细胞MMP-9基因表达比对照组高1.73倍(1.03-2.96);实验组第3d MMP-9基因表达比对照组高1.54倍(1.09-2.92);实验组第5d MMP-9基因表达比对照组高1.61倍(1.07-3.09)。实验组第1d与实验组第3、5d MMP-9表达ΔCT差异具有统计学意义(均P<0.05)。对照组第1、3、5d MMP-9组间基因表达ΔCT值差异具有统计学意义(均P<0.05),对照组第1d与对照组第3、5d的MMP-9表达差异具有统计学意义(ΔCT比较,均P<0.05)。ELISA结果显示实验组第1d、3d、5d MMP-9水平显著高于对照组同时间点MMP-9水平(均P<0.05)。对照组第3、5d血清MMP-9水平显著高于对照组第1d血清MMP-9水平(均P<0.05)。实验组第3、5d血清MMP-9水平显著高于实验组第1d血清MMP-9水平(均P<0.05)。结论 MMP-9在急性心肌梗死合并室颤发生、发展中起到促进作用,增加了急性心肌梗死患者发生室颤可能性。Objective To expolore the relevance of MMP-9 in acute myocardial infarction merger ventricular fibrillation. Methods 10 cases of acute myocardial infarction in our hospital patients with combination of ventricular fibrillation were enrolled as the experimental group;The same choice in 10 cases of acute myocardial infarction patients without combination of ventricular fibrillation were enrolled as control group. The experimental group and control group in incidence after 1,3,5d each extraction of peripheral venous blood 5ml,-20℃ refrigerator. By RT-PCR and elisa method and determine the MMP-9. Results RT-PCR re-sults show that the experimental group 1d cells MMP-9 gene expression was 1.73 times higher than the control group （1.03 2.96）;Group 3d MMP-9 gene expression was 1.54 times higher than the control group（1.09 2.92）;Group 5d MMP-9 gene expression was 1.61 times higher than the control group（1.07 3.09）. Experimental group 1d and 3,5d MMP-9ΔCT expression was differences sta-tistically significant（all P〈0.05）. Control group 1,3,5d gene expression between MMP-9 groups ΔCT value was differences statisti-cally significant （P〈0.05）,control group in 1d and control group in 3,5d of MMP-9 expression was differences statistically signifi-cant（ΔCT,P〈0.05）. Elisa results showed that the experimental group 1,3,5d level of MMP-9 was significantly higher than control group at the same time some level of MMP-9 （P〈0.05）. Control group in 3,5d was significantly higher serum levels of MMP-9 1d serum level of MMP-9 （P〈0.05）. Group 3,5d were significantly higher serum levels of MMP-9 experimental group 1d serum level of MMP-9 （P〈0.05）. Conclusion MMP-9 in acute myocardial infarction merger ventricular fibrillation play a promoting role in occurrence and development,increase the possibility ventricular fibrillation in patients with acute myocardial infarction.
