摘  要:[目的]研究桑黄菌丝体多糖对胶原诱导性关节炎(CIA)大鼠的治疗效果,并探讨其作用机制。[方法]采用水提醇沉法提取桑黄菌丝体多糖。将60只大鼠随机分为空白组、模型组、阳性组、桑黄菌丝体多糖低(100 mg·kg-1)、中(200 mg·kg-1)、高(400 mg·kg-1)剂量组,每组10只。给药期间采用足趾肿胀容积法测定大鼠非致炎足关节肿胀程度。30 d后麻醉大鼠,截取非致炎足做关节组织病理切片,显微镜下观察。ELISA法检测血清中白介素-17(IL-17)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-10(IL-10)的表达水平。[结果]与模型组比较,阳性对照组和桑黄菌丝体多糖各剂量组均能显著抑制非致炎足足趾肿胀(P<0.05),改善关节组织病理情况,降低IL-17、TNF-α、IL-1β的表达(P<0.05),升高IL-10的含量(P<0.05)。[结论]桑黄菌丝体多糖通过抑制炎症细胞因子IL-17、TNF-α、IL-1β的表达,促进抑炎因子IL-10的表达对CIA大鼠起到治疗作用。Objective]To explore the therapeutic effects of Phellinus igniarius mycelium polysaccharide on collagen-induced arxhritis(CIA) rats.[Methods] The method of water extract and alcohol precipitation was used to extract Phellinus igniarius mycelium polysaccharides.60 Wister rats were randomly divided into 6 groups,normal group,model group,control group,Phellinus igniarius mycelium polysaccharide high-dose,medium-dose and low-dose groups.The paw volume was measured to determine atrhroncus degree during administration.Ankle pathological sections were observed under microscope.The levels ofIL-17,TNF-α,IL-1 β,IL-10 were determined by ELISA.[Results] Each dose of Phellinus igniarius extracellular polysaccharide could decrease the degree of arthroncus(P〈0.05),improve joint tissue pathological situation,decrease the levels of TNF-α,IL-1β,IL-17(P〈0.05) and increase the level of IL-10(P〈0.05) significantly.[Conclusion] Phellinus igniarius mycelium polysaccharide may be through inhibiting the expression of irflammatory cytokines TNF-α,IL-1β,IL-17 and increasing the expression of inflammation suppression factor IL-10 to treat CIA rats.
