补肾活血通淋方对良性前列腺增生症模型大鼠Caspase-3的影响

摘　　要：目的:观察补肾活血通淋方对良性前列腺增生症大鼠模型Caspase-3基因的影响。方法:72只Wistar大鼠随机分为6组,即假手术组、模型组、保列治组、补肾活血通淋方低、中、高剂量组,除假手术组外,余组采用摘除大鼠双侧睾丸后连续注射丙酸睾丸酮30天的方法建立模型。在造模的同时,治疗组每天灌胃给药1次,连续30天,采用免疫组化法测定前列腺组织Caspase-3蛋白阳性平均灰度值,采用RTPCR法测定Caspase-3 mRNA的表达。结果:与模型组比较,各治疗组Caspase-3蛋白阳性平均灰度及mRNA相对表达量均明显升高(P<0.05),其中补肾活血通淋方高剂量组高于保列治组和补肾活血通淋方低剂量组(P<0.05)。结论:补肾活血通淋方可以上调大鼠良性前列腺增生症模型Caspase-3的表达,这可能是其治疗前列腺增生症的机制。This study was aimed to observe the effect of Bu-Shen Huo-Xue Tong-Lin （BSHXTL） formula on gene expression of Caspase-3 in benign prostatic hyperplasia （BPH） rats. A total of 72 rats were randomly divided into the sham operation group, model group, proscar group, low-dose BSHXTL group, middle-dose BSHXTL group and high- dose BSHXTL group. The rat model was established by injecting testosterone propionate for 30 days after removing testis except the sham operation group. The drugs were administered once a day at the same time of the model establishment. Immunohistochemical method was used to measure positive average gray. And RT-PCR was used to measure the gene expression of Caspase-3.The results showed that compared with the model group, the expression of Caspase-3 and mRNA in each treatment groups were increased （P 〈 0.05）. And the high-dose BSHXTL group was higher than the proscar group and low-dose BSHXTL group （P 〈 0.05）. It was concluded that BSHXTL formula may upregulate gene expression of Caspase-3, which may be the mechanism of BPH treatment.
