通塞脉片治疗2型糖尿病足模型大鼠的实验研究

摘　　要：目的探讨通塞脉片(TSM)对2型糖尿病足(DF)大鼠伤口愈合作用。方法用高脂高糖加链脲佐菌素(STZ)复制2型糖尿病大鼠,再行足部手术,建立DF大鼠模型。模型大鼠随机分为5组:模型组、二甲双胍0.135g/kg组、通塞脉片12.44、6.22、3.11g/kg 3个剂量组,另设空白对照组,连续灌胃给药18d,观察各组大鼠创面愈合率、血糖血脂、胰岛素、血液流变学以及血管内皮活性因子等指标的变化。结果 TSM可促进模型大鼠创面愈合、提高胰岛素敏感指数(ISI)、延长凝血酶原时间(PT)、凝血酶时间(TT)和活化部分凝血活酶时间(APTT),增加一氧化氮(NO)、一氧化氮合酶(NOS)、6-酮-前列腺素F1α(6-K-PGF1α)含量,降低模型大鼠血糖、血脂、胰岛素抵抗、血小板聚集率、血液黏度和内皮素(ET)、血栓素B2(TXB2),调节ET/NO、TXB2/6-K-PGF1α比值。结论 TSM可以通过调节糖脂代谢、减轻胰岛素抵抗、改善血液流变性、调节血管内皮细胞功能等作用,促进2型DF模型大鼠创面溃疡愈合。OBJECTIVE To investigate the effects of Tongsaimai （TSM） Tablet on rats with foot problems caused by type 2 diabetes. METHODS After rats were injected with STZ and then recieved foot surgery, SD rat model was established. The model rats were randomly divided into 5 groups, namely model group, 0. 135 g/kg metformin treatment group, 12.44 g/kg TSM treatment group, 6.22 g/kg TSM treatment group, 3.11 g/kg TSM treatment group and the control group. After intragastric 18 days administration to the rats, changes of wound healing rate, fasting blood glucose and lipid, insulin, hemorheology, and vascular endothelial active factors were observed. RESULTS For TSM groups, the wound healing rate and insulin sensitivity index （ISI） were improved, prothrombin time （PT）, thrombin time （TT） and activated partial thrombin time （APTT） were prolonged and nitric oxide （NO）, nitric oxide synthase （NOS） and 6-ketone-prostaglandinF1α in blood were increased, compared with model group （P〈0.05-0.01）. The fasting blood glucose （FBG）, blood lipid, insulin resistance index （HOMA-IR）, platelet aggregation rate （PAgT）, blood viscosity, endothelin （ET）, thromboxaneB2 （TXB2）, ET/NO, and TXB2/6-K-PGF1αof the TSM groups were significantly lower than that of the model group（P〈0.05-0.01）. CONCLUSION TSM tablet can promote the healing of ulcer of the rats with foot problems caused by type 2 diabetes by regulating the blood glucose and lipid functions of epithelial cells, reducing insulin resistance, and improving blood theology.
