摘 要:目的研究梓白皮、楸树皮和桑白皮对体外H2O2致损肝细胞的保护作用及其机制。方法采用H2O2(100μmol/L)体外诱导L-O2肝细胞损伤,CCK-8法检测细胞活性,检测细胞上清中天门冬氨酸转换酶(AST)、丙氨酸氨基转换酶(ALT),超氧化物歧化酶(SOD)和丙二醛(MDA)水平,ELISA法检测细胞中TNF-α和IL-1水平,流式细胞术检测细胞周期。结果 H2O2可以改变L-O2细胞的形态结构,抑制其活性;促进细胞代偿性增殖;提高细胞AST、ALT和TNF-α、IL-1的水平;降低细胞SOD活性。而梓白皮、楸树皮和桑白皮可拮抗H2O2对细胞的损伤。结论梓白皮、楸树皮和桑白皮对H2O2致损的LO2肝细胞有保护作用。OBJECTIVE To investigate the protective effect and mechanism of Bark of Chinese catalpa(Xinbaipi),bark of catalpa bungei(Jiushupi)and white mulberry root-bark(Sangbaipi)on damaged liver cell line L-O2 induced by H2O2 in vitro. METHODS Liver cell line L-O2 were incubated and then damaged by H2O2(100μmol/L)in vitro.The cell activity was detected by CCK-8 method.The levels of AST,ALT and MDA in cell supernatant were detected according to the kit.Levels of TNF-aand IL-1were detected by ELISA,and cell cycle was analyzed by flow cytometry.RESULTS H2O2 could change morphological structure of L-O2 cells,inhibit their activities,increase levels of AST,ALT,TNF-α and IL-1and inhibit activation of SOD in cells.However,bark of Chinese catalpa,bark of catalpa bungei and white mulberry root-bark could inhibit these injuries.CONCLUSION Bark of Chinese catalpa,bark of catalpa bungei and white mulberry root-bark can protect L-O2 cells from in vitro injuries induced by H2O2.
