敦化市2009-2011年致手足口病流行肠道病毒71型基因特征分析

摘　　要：目的分析吉林省敦化市流行的肠道病毒71型分离株的基因特征。方法对2009—2011年敦化市手足口病来源的3株EV71分离株进行逆转录一聚合酶链反应扩增VP1基因编码区后进行序列测定，并使用Bioedit和Maga4生物信息学软件进行基因特征分析。结果敦化市3株EV71流行株均为C4a基因亚型，3株EV71分离株之间核苷酸同源性为98．6％～98．8％；与吉林省和龙市的流行株核苷酸同源性为98．8％～99．8％，与吉林省其他地区的2株EV71代表株的核苷酸同源性为97．6％～99．3％，与安徽省阜阳市2008年代表株的核苷酸同源性为98．3％～99．3％。敦化市3株EV71分离株之间、与2008年阜阳株以及吉林省2009—2011年3株代表株的氨基酸同源性均达到100％。结论敦化市3株EV71分离株与同为延边州的和龙市的流行株核苷酸同源性最高，与安徽省阜阳市2008年流行株的核苷酸也具有较近的同源性。敦化市3株EV71分离株之间以及与中国C4a基因型代表株之间都具有较高的氨基酸同源性。Objective To analyze the genetic characteristics of enterovirus type 71 （EV71） strains isolated from suspected patients of Hand, Foot and Mouth Disease （HFMD） in Dunhua city of Jilin province. Methods The VP1 genes coding region of 3 EV71 strains from HFMD in Dunhua in 2009-2011 were amplified by reverse transcription-polymerase chain reaction （RT-PCR） and then sequenced. Gene characteristics were analyzed by Bioedit and Mega 4 software. Results All the 3 strains belonged to C4a subganotype.The homology of nncleotide sequences in VPI coding region of the 3 strains was 98.6%-98.8%; The homology of nucleotide sequences compared with those of Helong city, other region of Jilin province and Fuyang city were 98.8%-99.8%, 97.6%-99.3% and 98.3%-99.3% respectively. The homology of amino acid of them were 100%. Conclusions The 3 strains have the highest homology of nucleotide sequences compared with those of Helong strain, and also have the nearer homology of nucleotide sequences compared with Fuyang strain of 2008, The 3 strains have the higher homology of amino acid sequences compared with those of China representative strains of C4a.
