摘  要:【目的】研究单核细胞、内皮细胞的相互作用对单核细胞清道夫受体AI、CD36表达的影响,观察细胞因子在其中的介导作用。【方法】采用单核细胞U937、内皮细胞EA.hy926单独培养、隔离培养及混合培养的方法形成不同的培养条件,通过夹心酶联免疫吸附法检测培养液中巨噬细胞集落刺激因子的含量,使用流式细胞术检测单核细胞表面清道夫受体AI、CD36的表达程度。【结果】单核细胞单独培养条件下清道夫受体AI和CD36的表达量较低,在和内皮细胞隔离培养及混合培养后两者的表达量均显著性升高(P<0.05),巨噬细胞集落刺激因子和血小板源性生长因子BB在单核细胞清道夫受体AI和CD36表达增高的调节中起着一定的介导作用,但不占主要地位。【结论】单核细胞与内皮细胞的相互作用可通过多种环节上调单核细胞清道夫受体AI和CD36的表达,从而参与动脉粥样硬化的发展。[Objective]To examine the effect of monocyte-endothelium interaction on the expression of scavenger receptor AI(SRAI) and CD36 in monocytes and research the functions of cytokines in this process, [Methods] Monocyte strain U937 and endothelial cell strain EA.hy926 were cultured alone, cocuhured and isolatedly cocultured in order to form different cell culture conditions. The level of macrophage colony-stimulating factor in culture medium was determined by enzyme linked immune sandwich assay technique, and the expression of SRAI and CD36 in monocytes was determined by flow cytometry. [ Results] The expression of AI and CD36 was lower in monocytes was low in monocytes cultured alone but increased significantly when monocytes and endothelial cells were cocultured or iso- lated isolatedly cocultured (P 〈 0.05). Macrophage colony-stimulating factor and platelet derived growth factor BB played certain roles in increased expression of SRAI and CD36 in monocytes, but they didn't seize the principal position. [Conclusion ] The monocyte-en- dothlium interaction could increase the expression of SRAI and CD36 in monocytes through various mechanisms and might participate in the pathogenesis of atherosclerosis.
